| 目的 探讨前列腺肿瘤过表达基因1(PTOV1)在I型子宫内膜癌组织中检测的意义及表达调控的分子机制。方法收集本院住院治疗的46例I型子宫内膜癌患者手术切除肿瘤组织及癌旁组织,免疫组化法检测组织中PTOV1蛋白的表达,比较瘤组织与癌旁组织的差异,亚硫酸氢盐测序法检测PTOV1启动子甲基化状态,比较瘤组织与癌旁组织的差异,统计分析PTOV1蛋白表达与I型子宫内膜癌患者临床参数的关系,PTOV1启动子甲基化与蛋白表达的关系及其与患者临床参数的关系。结果PTOV1在I型子宫内膜癌组织中高表达且启动子低甲基化；PTOV1高表达与I型子宫内膜癌患者年龄、肿瘤大小、组织分化程度、淋巴结转移无关,与浸润深度呈正相关；PTOV1启动子低甲基化与I型子宫内膜癌组织中PTOV1表达呈正相关。结论PTOV1与I型子宫内膜癌浸润密切相关,其高表达的机制可能为启动子发生低甲基化。 |

| Objective To identify the significance of detecting prostate tumor overexpressed 1(PTOV1) and the mechanism of its expression regulation in type I endometrial cancer.MethodsForty-six tumor tissues and para-cancer tissues from typeIendometrial cancer patients were collected.The expression of PTOV1 protein in typeIendometrial cancer tissues and para-cancer tissues was detected by immunohistochemistry.The methylation status of PTOV1 was tested by bisulfite sequencing PCR.The differences between cancer tissues and para-cancer tissues were analyzed.The correlation between the PTOV1 expression and clinical parameters,the methylation status of PTOV1 and its expression,and the methylation status of PTOV1 and clinical parameters was statistically analyzed.ResultsThe expression of PTOV1 was increased and promotermethylation status was decreased in typeIendometrial cancer tissues.There was no correlation between PTOV1 expression and age,tumor size,differentiation degree,lymph node metastasis.But PTOV1 expression was positively related to invasion depth.Hypomethylation of PTOV1 promoter was positively related to its expression in type I endometrial cancer.ConclusionPTOV1 expression was correlated with typeIendometrial cancer invasion depth.The mechanism of PTOV1 expression may be promoter hypomethylation. |
