| 目的观察罗格列酮对去卵巢大鼠学习记忆功能及海马内肿瘤坏死因子-α（TNF-α）和白介素-6（IL-6）表达的影响。方法56只SD雌性大鼠，随机分为4组：假手术对照组、去卵巢组、假手术+罗格列酮组、去卵巢＋罗格列酮治疗组，第15周进行Morris水迷宫试验。第16周处死大鼠，ELISA法检测大鼠大脑海马区的TNF-α、IL-6的表达。结果与假手术组比较，去卵巢组大鼠学习记忆能力明显下降（P＜0.01），大脑海马区TNF-α和IL-6的表达明显升高（均P＜0.01）；与去卵巢组比较，罗格列酮治疗组大鼠学习记忆能力明显改善，大脑海马区TNF-α和IL-6的表达明显降低（均P＜0.01）。结论罗格列酮能改善去卵巢大鼠的学习记忆障碍，其机制可能与其降低海马TNF-α和IL-6的表达有关。 |

| ObjectiveTo investigate the effect of rosiglitazone on the learning and memory and expression of tumor necrosis factor-α and interleukin-6 in the cerebrum in ovariectomized rats.MethodsFifty-six male Sprague-Dawley rats were randomly divided into 4 groups:the sham control group (SC),ovariectomized group (OV),sham control +rosiglitazone group (SC+R) and ovariectomized +rosiglitazone group,(OV+R).The Morris water maze test was performed at the fifteenth week.The rats were sacrificed at the sixteenth week.The expressions of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in the hippocampus were measured by ELISA assay.ResultsCompared with the SC group,the ability of learning and memory were significantly decreased and the expressions of TNF-α and IL-6 in the hippocampus were significantly increased in the OV group (all P＜0.01).Compared the OV group,the ability of memory was significantly increased and the expressions of TNF-6 and IL-6 in the hippocampus were significantly decreased in the OV +R group (all P＜0.01).ConclusionRosiglitazone improves the impairment of learning and memory in ovariectomized rats,the mechanism of which may be associated with the down-regulation of TNF-α and IL-6 in the hippocampus induced by rosiglitazone. |
