      目的建立反相高效液相色谱法测定人血清中茶碱浓度。方法血清样品加入咖啡因为内标,10%三氯醋酸去蛋白,采用Ultimate XB-C18(250 mm×4.6 mm,5 μm)色谱柱分离,流动相为甲醇∶水=30∶70,流速为1.0 mL/min,检测波长为273 nm,柱温为30℃。结果茶碱在1.76~44.00 mg/L浓度范围线性关系良好,r=0.999 6,血清最低检测限为0.44 mg/L,日内精密度和日间精密度良好,回收率较高。结论本法操作简便,结果准确可靠。可满足临床快速监测的需要。

      ObjectiveTo determine theophylline rapidly in serum by concentration of theophylline in serum.MethodThe separation was performed on Ultimate XB-C18(4.6 mm×250 mm,5 μm) column with methanol-water( 30∶70 ) as mobile phase.The flow rate was 1.0 mL/min.The wavelength for detection was 273 nm.The column temperature was 30℃.Trichloroacetic acid was used to precipitate the serum protein.ResultsThe calibration curve had good linearity between 1.76~44.00 mg/L,r=0.999 6.The intra-day precision and inter-day precision were all good.The average recovery were all respective.ConclusionsThis method is accuate,rapid,and simple.It can be used as a roution method by TDM.
