      【】 目的 观察黄芪对慢性哮喘豚鼠模型气道重塑的研究。方法 SPF级雄性豚鼠40只随机分成4组:正常对照组 (A组) ,模型组(B组),黄芪注射液组 ( C组),地塞米松组(D组)每组10只。采取HE染色观察气道重塑,图像分析仪分析肺组织气道重塑情况,免疫组化方法测气道中TGF-β1蛋白的表达。结果 1、豚鼠肺组织病理切片显示:B组气道壁结构发生改变,气道平滑肌增殖,支气管管腔变窄,气道上皮细胞脱落,平滑肌增生肥大。C组和D组经治疗后,其改变较哮喘组减轻,A组无上述改变。2、图像分析结果表明,哮喘组支气管壁厚度(WA/Pi)、支气管壁平滑肌厚度(平滑肌的面积/Pi)、支气管壁平滑肌细胞核数量(N/Pi)分别较对照组显著增加。3、免疫组化蛋白半定量分析,D组TGF-β1蛋白的积分光密度值(IOD)均较A组明显增高,B、C组IOD值介于两者之间。结论 1.哮喘豚鼠TGF-β1表达水平增高, TGF-β1可能参与了哮喘大鼠的气道重塑。2.黄芪注射液能够抑制慢性哮喘模型的气道重塑,其机制可能通过抑制TGF-β1的表达而实现的。

      【】 Objective To investigate the expression of TGF-β1 in the airway epithelial cell of asthmatic guinea pigs and study the effects of Huang Qi on airway remolding in asthma .Methods Forty adult male guinea pigs were randomly divided into control group (group A),asthmatic group (group B), arsenic huang qi group (group C) and dexamethasone group(group D). Use HE staining and the image analysis device to analyse the information of the airway remodeling. Immunohistochemistry detected the expression of TGF-β1 in the lung tissue. Results 1.Lung pathological sections of Group B showed proliferation of airway smooth muscle , reconstruction of air passage ,narrowing of bronchial lumen and swelling of airway epithelial cells. After the treatment,The pathologic changes of group C and D were milder than in group B .Group A is without the above change,the differences were significant (P < 0.01). 2.The results of image analysis showed that the airway wall and smooth muscle layer were much thicker in asthma group than those of control group, the number of smooth muscle cell nucleus increased significantly, the differences were significant (P < 0.01).3.Immunohistochemical semi-quantitative analysis showed integrated optical density (IOD) value of Group D were statistically significantly higher than that in group A,the differences were significant (P < 0.01), the IOD values in B and C group is situated between the two.Conclusions 1.There was TGF-β1- overproduction in airway epithelial cells of asthmatic guinea-pigs, which may contribute to airway remolding in asthma.2 Astragus could decrease airway remolding by inhibiting the expression of TGF-β1.
