| 本文旨在研究长链非编码RNA UNC5B-AS1在胃癌中差异表达的临床意义及其对胃癌细胞活力和迁徙的影响。从TCGA和GEO数据库中筛选出所有差异表达的mRNAs和lncRNAs,通过qRT-PCR检测mRNA水平UNC5B-AS1在胃癌中的差异表达；CCK-8实验检测UNC5B-AS1表达对胃癌细胞增殖的影响；划痕实验检测UNC5B-AS1对胃癌细胞迁移的影响。结果显示,相较于癌旁组织,UNC5B-AS1在人胃癌组织中表达显著下调,但胃癌患者的总生存期无显著影响,高表达UNC5B-AS1的胃癌患者无复发生存期较差,且相对于Ⅰ期胃癌患者,Ⅱ-Ⅳ期的胃癌患者高表达UNC5B-AS1,异常表达的UNC5B-AS1对胃癌细胞的增殖无显著影响,然而敲降UNC5B-AS1,胃癌细胞的迁移能力显著增加；过表达UNC5B-AS1,胃癌细胞的迁移能力显著抑制。结果推测UNC5B-AS1可作为评价胃癌复发和转移的相关标志物和潜在的治疗靶点。 |

| The aim of this study was to investigate the clinical significance of differential expression of long non-coding RNAs (lncRNAs) UNC5B-AS1 in gastric cancer, and to analyze its impact on the function of gastric cancer cells. Screening all differentially expressed RNAs and lncRNAs from TCGA and GEO databases. Detection of differential expression of UNC5B-AS1 in gastric cancer by qRT-PCR. The effect of UNC5B-AS1 on the proliferation of gastric cancer cells was detected by CCK-8 assay. The effect of UNC5B-AS1 on the migration of gastric cancer cells was detected by scratch assay. Results showed, compared with adjacent normal tissues, UNC5B-AS1 was significantly down-regulated expression in human gastric cancer tissues,but the overall survival of patients with gastric cancer was not significantly affected.Gastric cancer patients with high expression of UNC5B-AS1 have poorer recurrence-free survival, and UNC5B-AS1 was highly expressed in stage Ⅱ-Ⅳ tissues than stage Ⅰ tissues.Abnormally expressed UNC5B-AS1 had no significant effect on the proliferation of gastric cancer cells. However, knockdown of UNC5B-AS1 significantly increased the migration of gastric cancer cells, and overexpression of UNC5B-AS1 significantly inhibited the migration of gastric cancer cells. UNC5B-AS1 can be used as a marker and potential therapeutic target for evaluate recurrence and metastasis of gastric cancer. |
