| 目的分析结直肠癌患者CD8+T细胞浸润密度、肿瘤免疫逃逸因子表达及其预后影响因素。 方法选取130例结直肠癌患者的结直肠癌组织及配对的正常组织。采用免疫组化染色评估CD8+T细胞浸润密度,检测肿瘤微环境中免疫逃逸因子[程序性死亡受体-1(PD-1)、程序性死亡受体配体-1(PD-L1)]表达情况,并通过单因素和多因素COX回归分析患者预后的影响因素。 结果结直肠癌组织中CD8+T细胞浸润密度低于癌旁正常组织(P<0.05);肿瘤免疫逃逸因子PD-1和PD-L1的表达率均高于癌旁正常组织(P<0.05)。Pearson相关性分析结果显示,PD-1和PD-L1表达率与CD8+T细胞浸润密度呈负相关(P<0.05)。COX多因素回归分析结果显示,TNM分期、肿瘤分化程度、CD8+T细胞浸润密度及PD-1、PD-L1表达率为结直肠癌预后的影响因素(P<0.05)。 结论结直肠癌组织中CD8+T细胞浸润与肿瘤免疫逃逸机制密切相关。TNM分期、肿瘤分化程度、CD8+T细胞浸润密度、PD-1、PD-L1表达率为结直肠癌患者预后的影响因素。 |

| AimTo analyze the infiltration density of CD8+ T cells, the expression of tumor immune escape factors and the prognostic influencing factors in patients with colorectal cancer. MethodsColorectal cancer tissues and paired normal tissues were collected from 130 patients with colorectal cancer. Immunohistochemical staining was used to evaluate the infiltration density of CD8+ T cells and detect the expression of immune escape factors [programmed death-1 (PD-1), programmed death-ligand 1 (PD-L1)] in the tumor microenvironment. Univariate and multivariate COX regression analyses were performed to identify the influencing factors of patient prognosis. ResultsThe infiltration density of CD8+ T cells in colorectal cancer tissues was significantly lower than that in adjacent normal tissues (P<0.05); The expression rates of tumor immune escape factors PD-1 and PD-L1 in colorectal cancer tissues were significantly higher than those in adjacent normal tissues (P<0.05). Pearson correlation analysis showed that the expression rates of PD-1 and PD-L1 were negatively correlated with the infiltration density of CD8+T cells (P<0.05). Multivariate COX regression analysis indicated that TNM stage, tumor differentiation degree, CD8+ T cell infiltration density, and PD-1, PD-L1 expression rates were influencing factors for the prognosis of colorectal cancer (P<0.05). ConclusionThe infiltration of CD8+ T cells in colorectal cancer tissues is closely related to the mechanism of tumor immune escape. TNM stage, tumor differentiation degree, CD8+T cell infiltration density, and PD-1, PD-L1 expression rates are the influencing factors for the prognosis of patients with colorectal cancer. |
