      目的探讨环氧合酶-2(COX-2)抑制剂尼美舒利对肝星状细胞(HSC)-T6细胞增殖、凋亡的影响。 方法将HSC-T6细胞实验分为对照组(RPMI-1640培养液)、低剂量组(尼美舒利200 μmol/L培养液)和高剂量组(尼美舒利400 μmol/L培养液)。采用免疫细胞化学法检测各组COX-2蛋白表达水平,MTT法检测各组细胞增殖能力,流式细胞术检测各组细胞凋亡情况。 结果低剂量组、高剂量组COX-2表达水平均低于对照组(P<0.05)。在48、72 h时,HSC-T6细胞增殖能力为高剂量组<低剂量组<对照组(P<0.05)。高剂量组、低剂量组HSC-T6细胞凋亡率均高于对照组(P<0.05)。 结论尼美舒利对大鼠HSC-T6细胞具有抑制增殖、诱导凋亡的作用。

      AimTo explore the effects of cyclooxygenase-2 (COX-2) inhibitor nimesulide on the proliferation and apoptosis of hepatic stellate cell(HSC)-T6. MethodsThe HSC-T6 cells experiment were divided into a control group (RPMI-1640 medium), a low-dose group (nimesulide 200 μmol/L medium), and a high-dose group (nimesulide 400 μmol/L medium). Immunocytochemistry was used to detect the expression level of COX-2 protein in each group, MTT assay was used to detect the cell proliferation ability of each group, and flow cytometry was used to detect cell apoptosis. ResultsThe expression levels of COX-2 in both low-dose and high-dose groups was lower than that in the control group (P<0.05). At 48 and 72 hours, the proliferation ability of HSC-T6 cells was in an order of the high-dose group<the low-dose group<the control group (P<0.05). The apoptosis rate of HSC-T6 cells in the high-dose and low-dose groups was higher than that in the control group (P<0.05). ConclusionNimesulide has inhibitory effects on proliferation and induces apoptosis in rat HSC-T6 cells.
