| 目的分析丹酚酸B通过Toll样受体4(TLR4)/核因子-κB(NF-κB)信号通路在大鼠心肌细胞缺氧复氧损伤修复中的作用。 方法将大鼠心肌细胞H9c2分为正常组、模型组、丹酚酸B组(6.25 mg/L)和激活剂组(6.25 mg/L丹酚酸B+15 μmol/L TLR4激活剂)。MTT法检测细胞活性,流式细胞仪检测细胞凋亡率,采用试剂盒检测细胞乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)、丙二醛(MDA)、白细胞介素(IL)-1β、IL-6水平,蛋白免疫印迹法检测B细胞淋巴瘤2(Bcl-2)、凋亡调控因子Bcl-2相关X蛋白(Bax)、TLR4、NF-κB p65、p-NF-κB p65蛋白水平。 结果模型组细胞存活率、SOD、Bcl-2蛋白、Bcl/Bax低于正常组,丹酚酸B组高于模型组,激活剂组低于丹酚酸B组(P<0.05)。模型组细胞凋亡率、MDA、LDH活性、IL-1β、TNF-α、IL-6、Bax蛋白、TLR4、p-NF-κB p65/NF-κB p65蛋白水平高于正常组,丹酚酸B组低于模型组,激活剂组高于丹酚酸B组(P<0.05)。 结论丹酚酸B可提高缺氧复氧损伤心肌细胞存活率、降低凋亡率,抑制炎症反应和氧化应激,其保护作用可能是通过抑制TLR4/NF-κB信号通路实现的。 |

| AimTo analyze the role of salvianolic acid B in the repairing of hypoxia-reoxygenation injury in rat cardiomyocytes via the Toll-like receptor 4 (TLR4)/nuclear factor-κB (NF-κB) signaling pathway. MethodsH9c2 rat cardiomyocytes were divided into normal group, model group, salvianolic acid B group (6.25 mg/L), and activator group (6.25 mg/L salvianolic acid B+15 μmol/L TLR4 activator). MTT assay was used to detect cell viability, flow cytometry was used to detect cell apoptosis rate, and assay kits were used to measure the levels of lactate dehydrogenase (LDH), superoxide dismutase (SOD), malondialdehyde (MDA), interleukin-1β and IL-6. Protein immunoblotting was used to detect the protein levels of B cell lymphoma 2 (Bcl-2), apoptosis regulatory factors Bcl-2 associated X protein (Bax), TLR4, NF-κB p65, and p-NF-κB p65. ResultsThe cell survival rate, SOD, Bcl-2 protein, and Bcl/Bax in the model group were lower than those in the normal group, higher in the salvianolic acid B group than those in the model group, and lower in the activator group than those in the salvianolic acid B group (P<0.05). The apoptosis rate, MDA, LDH activity, IL-1β, TNF-α, IL-6, Bax protein, TLR4, p-NF-κB p65/NF-κB p65 protein levels in the model group were higher than those in the normal group, lower in the salvianolic acid B group than those in the model group, and higher in the activator group than those in the salvianolic acid B group (P<0.05). ConclusionDanshensu B can increase the survival rate of myocardial cells under hypoxia reoxygenation injury, reduce apoptosis rate, and inhibit inflammatory response and oxidative stress.Its protective effect may be achieved by inhibiting the TLR4/NF-κB signaling pathway. |
