      目的探讨针刺联合康复训练对缺血性脑卒中小鼠脑血流量及腺苷酸活化蛋白激酶(AMPK)/谷氨酰胺-6-磷酸果糖转氨酶-1(GFAT-1)/血管内皮生长因子(VEGF)信号通路的影响。 方法将60只小鼠随机均分为假手术组、模型组、康复组、针刺组和联合组(康复+针刺)。除假手术组外,其余组构建缺血性脑卒中模型。干预14天后,采用Longa评分评估神经功能,激光散斑血流成像仪检测脑血流灌注量,HE染色观察脑组织病理变化,TTC染色评估脑梗死体积,酶联免疫吸附法检测白细胞介素(IL)-6、IL-1β及肿瘤坏死因子-α(TNF-α)水平,蛋白免疫印迹法检测AMPK、磷酸化AMPK(p-AMPK)、GFAT-1及VEGF蛋白水平。 结果与假手术组比较,其他组脑神经功能Longa评分、脑梗死体积、IL-1β、IL-6、TNF-α、GFAT-1蛋白水平升高,脑血流灌注量、AMPK、p-AMPK、VEGF蛋白水平降低(P<0.05);与模型组比较,康复组、针刺组和联合组上述趋势得到逆转,且联合组改善优于康复组、针刺组(P<0.05)。模型组脑组织细胞排列不规则,细胞间隙变大,可见细胞坏死;康复组、针刺组和联合组小鼠脑组织病理改变明显改善,且联合组改善最显著。 结论针刺联合康复训练可通过调控AMPK/GFAT-1/VEGF信号通路改善缺血性脑卒中小鼠神经损伤,保护脑组织。

      AimTo investigate the effects of acupuncture combined with rehabilitation training on cerebral blood flow and the adenosine monophosphate-activated protein kinase (AMPK)/glutamine-fructose-6-phosphate aminotransferase-1 (GFAT-1)/vascular endothelial growth factor (VEGF) signaling pathway in mice with ischemic stroke. MethodsSixty mice were randomly divided into sham, model, rehabilitation, acupuncture, and combined (rehabilitation + acupuncture) groups, with 12 mice in each group. Except the sham group, the other groups were subjected to ischemic stroke modeling. After 14 days of intervention, neurological function was assessed using the Longa score, cerebral blood flow perfusion was measured by laser speckle imaging, brain tissue pathology was observed by hematoxylin-eosin (HE) staining, cerebral infarction volume was evaluated by triphenyltetrazolium chloride (TTC) staining, interleukin (IL)-6, IL-1β, and tumor necrosis factor-α (TNF-α) levels were detected by enzyme-linked immunosorbent assay, and AMPK, phosphorylated AMPK (p-AMPK), GFAT-1, and VEGF protein levels were measured by Western blotting. ResultsCompared with the sham group, the Longa score, cerebral infarction volume, IL-1β, IL-6, TNF-α, and GFAT-1 protein levels were increased, while cerebral blood flow perfusion, AMPK, p-AMPK, and VEGF protein levels were decreased in the other groups (P<0.05). Compared with the model group, the rehabilitation, acupuncture, and combined groups showed significant improvements in these indicators, with the combined group showing the most significant improvement (P<0.05). The model group exhibited disordered cell arrangement, enlarged intercellular spaces, and necrosis, while the rehabilitation, acupuncture, and combined groups showed significant pathological improvements, with the combined group showing the best results. ConclusionAcupuncture combined with rehabilitation training can improve neurological damage and protect brain tissue in mice with ischemic stroke by regulating the AMPK/GFAT-1/VEGF signaling pathway.
