目的探讨miR-1247-3p对子宫内膜癌肺转移的影响机制。 方法将Ishikawa细胞实验分为短发夹CARD14(shCARD14)组、shCARD14对照(shCARD14-NC)组、miR-1247-3p抑制剂(i-miR-1247-3p)组、i-miR-1247-3p-NC组、Control组、miR-1247-3p-NC组、miR-1247-3p组、shCARD14+miR-1247-3p组。CCK-8、划痕及Transwell实验检测各组细胞增殖、迁移及侵袭能力。Western blotting检测各组CARD14、核因子-κB(NF-κB)通路相关蛋白的表达水平。流式细胞术检测各组细胞周期。双荧光素酶报告实验检测miR-1247-3p与CARD14的结合作用。在裸鼠实验中,通过尾静脉注射建立裸鼠肺转移模型,对肺转移结节进行计数,并通过HE染色观察肺组织的形态学特征。 结果miR-1247-3p通过调控CARD14基因表达水平,抑制体外癌细胞增殖、迁移及侵袭能力,下调NF-κB通路相关蛋白的表达水平。miR-1247-3p与CARD14存在结合位点,miR-1247-3p负调控CARD14的表达(P<0.05)。裸鼠验证实验结果显示,miR-1247-3p抑制肺转移,降低肺转移结节数量、NF-κB相关蛋白以及CARD14蛋白表达水平,并改善肺组织形态学特征。 结论miR-1247-3p在子宫内膜癌转移过程中通过下调CARD14的表达,抑制NF-κB通路激活,从而抑制子宫内膜癌肺转移。 |

AimTo explore the mechanism of miR-1247-3p on lung metastasis in endometrial cancer. MethodsThe Ishikawa cell experiment was divided into short hairpin CARD14 (shCARD14) group, shCARD14 control (shCARD14-NC) group, miR-1247-3p inhibitor (i-miR-1247-3p) group, i-miR-1247-3p-NC group, Control group, miR-1247-3p-NC group, miR-1247-3p group, and shCARD14+miR-1247-3p group. Cell proliferation, migration, and invasion abilities were assessed by using CCK-8, scratch wound assays, and Transwell assays, respectively. Western blotting was conducted to detect the expression levels of CARD14 and proteins in the nuclear factor-κB (NF-κB) signaling pathway in Ishikawa cells, and flow cytometry was used to analyze the cell cycle. Dual-luciferase reporter assays were performed to confirm the interaction between miR-1247-3p and CARD14. In the nude mice experiments, a nude mice lung metastasis model was established by tail vein injection, and lung metastatic nodules were counted.Lung tissue morphology was examined by using HE staining. ResultsmiR-1247-3p inhibited the proliferation, migration, and invasion of cancer cells in vitro, and downregulated the expression of related proteins in the NF-κB pathway by regulating CARD14 gene expression level. miR-1247-3p had a binding site with CARD14, and miR-1247-3p negatively regulated the expression of CARD14 (P<0.05). The results of nude mouse validation experiments showed that miR-1247-3p significantly inhibited lung metastasis, reduced the number of lung metastatic nodules and protein expression levels of NF-κB related proteins and CARD14 protein, and improved the morphological characteristics of lung tissue. ConclusionmiR-1247-3p inhibits lung metastasis of endometrial cancer by downregulating CARD14 gene expression and suppressing the activation of the NF-κB signaling pathway. |
