      目的研究高脂饮食对新西兰兔脑组织细胞凋亡及枯草溶菌素转化酶9(PCSK9)表达水平的影响。 方法将12月龄健康纯种新西兰兔12只随机均分为普食组(普通饲料喂养)和高脂组(高胆固醇饲料喂养)。比较两组血浆甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白(LDL)和高密度脂蛋白(HDL)含量;HE染色及油红O染色检测大脑和小脑病理形态学改变及脂质蓄积情况;Hoechst 33258染色和PI染色流式细胞术检测大脑和小脑组织细胞凋亡情况;RT-PCR检测大脑、小脑组织LDL受体(LDLR)mRNA表达水平;Western blotting检测大脑组织PCSK9蛋白的表达水平。 结果HE染色结果显示,高脂组脑组织出现明显病理学改变;油红O染色结果显示高脂组脑组织脂质蓄积高于普食组。高脂组新西兰兔血浆TG、TC、LDL、HDL含量、大脑组织PCSK9表达水平、小脑组织细胞凋亡率明显高于普食组,而脑组织LDLR mRNA表达水平明显低于普食组(P<0.05)。结论高脂饮食促进新西兰兔脑组织细胞凋亡,可能与PCSK9蛋白表达增加引起的神经退行性疾病有关。

      AimTo study the effects of high-fat diet on brain tissue cell apoptosis and PCSK9 expression levels in New Zealand rabbits. Methods12 healthy purebred New Zealand rabbits of 12-month-old were randomly divided into the normal diet group (fed with normal diet) and the high-fat diet group (fed with high cholesterol diet). The levels of plasma triglyceride (TG), total cholesterol (TC), low density lipoprotein (LDL) and high density lipoprotein (HDL) were compared between the two groups. HE staining and oil red O staining were used to detect the pathological changes and lipid accumulation in the cerebrum and cerebellum.Hoechst 33258 staining and PI staining flow cytometry were used to detect the apoptosis of cerebrum and cerebellar tissues. RT-PCR was used to detect the expression level of LDL receptor (LDLR) mRNA in brain and cerebellar tissues. Western blotting was used to detect the expression level of PCSK9 protein in brain tissue. ResultsHE staining results showed that there were significant pathological changes in brain tissue of high-fat diet group. Oil red O staining results showed that lipid accumulation in brain tissue of high-fat diet group was higher than that of normal diet group. The contents of TG, TC, LDL, HDL in plasma of New Zealand rabbits, the expression of PCSK9 protein in cerebrum tissue, and the apoptosis rate of cerebellar tissue cells in high-fat diet group were significantly higher than those in normal diet group, while the expression of LDLR mRNA in brain tissue was significantly lower than that in normal diet group (P<0.05). ConclusionHigh-fat diet promotes apoptosis of brain tissue cells in New Zealand rabbits, which may be related to neurodegenerative diseases caused by increased expression of PCSK9 protein.
