目的研究姜黄素通过调控miR-221对6-羟基多巴胺(6-OHDA)诱导帕金森病(PD)细胞模型氧化损伤的影响。 方法PC12细胞分为正常对照组、PD组、姜黄素组;PD细胞转染后分为PD+miR-NC组和PD+miR-221组,以及PD+anti-miR-NC+姜黄素组和PD+anti-miR-221+姜黄素组。通过流式细胞术检测细胞凋亡;Western blotting检测Caspase-3蛋白表达;比色法分析活性氧(ROS)水平,分光光度计法测定超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量;qRT-PCR检测细胞miR-221表达水平。 结果与正常对照组比较,PD组细胞凋亡率、Caspase-3蛋白表达、ROS水平及MDA含量升高,SOD活性、miR-221水平降低(P<0.05);与PD组比较,姜黄素组细胞凋亡率、Caspase-3蛋白表达、ROS水平及MDA含量降低,SOD活性、miR-221水平升高(P<0.05);与PD+miR-NC组比较,PD+miR-221组凋亡率、Caspase-3蛋白表达、ROS水平及MDA含量降低,SOD活性、miR-221水平升高(P<0.05);与PD+anti-miR-NC+姜黄素组比较,PD+anti-miR-221+姜黄素组凋亡率、Caspase-3表达、ROS水平及MDA含量升高,SOD活性、miR-221水平降低(P<0.05)。 结论姜黄素上调miR-221可减轻6-OHDA诱导的PD细胞模型氧化损伤。 |

AimTo study the effect of curcumin on 6-hydroxydopamine (6-OHDA)-induced oxidative damage in Parkinson (PD) cell model by regulating miR-221. MethodsPC12 cells were divided into normal control group, PD group, and curcumin group. After transfection, PD cells were further divided into PD+miR-NC group and PD+miR-221 group, PD+anti-miR-NC+curcumin group and PD+anti-miR-221+curcumin group. Apoptosis was detected by flow cytometry; the expression of Caspase-3 protein was detected by Western blotting; the levels of reactive oxygen species (ROS) were determined by colorimetry, and the activity of superoxide dismutase (SOD) and the content of malondialdehyde (MDA) were determined by spectrophotometer; qRT-PCR was used to detect the expression level of miR-221 in cells. ResultsCompared with normal control group, apoptosis rate, expression of Caspase-3 protein, level of ROS and content of MDA were increased in PD group, while activity of SOD and level of miR-221 were decreased (P<0.05). Compared with PD group, the apoptosis rate, apoptosis rate, expression of Caspase-3 protein, level of ROS and content of MDA in curcumin group were decreased, while activity of SOD and level of miR-221 were increased (P<0.05). Compared with PD+miR-NC group, apoptosis rate, expression of Caspase-3 protein, level of ROS and content of MDA in PD+miR-221 group were decreased, and activity of SOD and level of miR-221 was increased (P<0.05). Compared with PD+anti-miR-NC+curcumin group, apoptosis rate, expression of Caspase-3 protein, ROS level and MDA content in PD+anti-miR-221+curcumin group were increased, while activity of SOD and level of miR-221 was decreased (P<0.05). ConclusionCurcumin can reduce 6-OHDA-induced oxidative damage in PD cell models by up-regulating miR-221. |
