      目的探究缺氧缺血性脑损伤对新生小鼠脑组织水通道蛋白(AQP)-9表达和空间学习记忆能力的影响。 方法阻断子宫动脉怀孕雌鼠所产小鼠为缺氧缺血性脑病(HIE)新生小鼠,纳入HIE组;不阻断子宫动脉怀孕雌鼠所产小鼠纳入对照组。比较两组小鼠神经元分布情况。采用免疫组化法检测小鼠海马CA1区、CA3区和DG区小清蛋白的表达。采用Western blotting检测小鼠海马神经传递相关蛋白NR2B、EAAT1、AQP-9及谷氨酸脱羧酶(GAD)的表达。采用新事物识别实验检测小鼠空间学习记忆能力。 结果与对照组比较,HIE组CA1区、CA3区神经元密度减少(P<0.05),但与DG区神经元密度差异无显著性(P>0.05)。HIE组3个区域小清蛋白表达均高于对照组(P<0.05)。HIE组AQP-9、GAD表达水平高于对照组(P<0.05)。HIE组小鼠存在空间学习记忆缺陷。 结论HIE新生小鼠脑组织AQP-9表达水平升高,空间学习记忆能力降低。
英文摘要:
      AimTo explore the effects of hypoxic ischemic brain injury on the expression of AQP-9 and spatial learning and memory abilities in the brain tissue of neonatal mice. MethodsNeonatal mice with hypoxic-ischemic encephalopathy (HIE) born from pregnant mice with blocked uterine arteries were included in the HIE group; Mice born from pregnant female mice without blocking uterine arteries were included in the control group. The distribution of neurons between two groups was compared. Immunohistochemical method was used to detect the expression of small albumin in the CA1, CA3, and DG regions of mouse hippocampus. Western blotting was used to detect the expression of neurotransmission-related proteins NR2B, EAAT1, AQP-9, and glutamic acid decarboxylase (GAD) in the hippocampus of mice. New object recognition experiments were used to test the spatial learning and memory abilities of mice. ResultsCompared with the control group, the neuron density in the CA1 and CA3 regions of the HIE group were decreased (P<0.05), but this change showed no significant difference compared with the DG region (P>0.05). The expression of small albumin in all three regions of the HIE group was higher than that of the control group (P<0.05). The expression levels of AQP-9 and GAD in the HIE group were higher than those in the control group (P<0.05). The HIE group mice have spatial learning and memory deficits. ConclusionThe expression level of AQP-9 in the brain tissue of HIE newborn mice increases, and their spatial learning and memory abilities decrease.
