目的探讨circ_0000502对甲状腺乳头状癌TPC-1细胞增殖、迁移、侵袭和凋亡的影响。 方法选取26例甲状腺乳头状癌组织及相应癌旁组织。将TPC-1细胞分为NC组、si-NC组、si-circ_0000502组、miR-NC组、miR-1231组、si-circ_0000502+anti-miR-NC组、si-circ_0000502+anti-miR-1231组。qRT-PCR检测circ_0000502和miR-1231的表达水平;MTT检测细胞活性;Transwell、流式细胞术检测细胞的迁移、侵袭和细胞凋亡情况;Western blotting检测增殖细胞核抗原(PCNA)、基质金属蛋白酶(MMP)-2、MMP-9、B淋巴细胞瘤-2(Bcl-2)、Bcl-2相关X蛋白(Bax)蛋白表达;双荧光素酶报告基因实验检测circ_0000502和miR-1231的靶向关系。 结果甲状腺乳头状癌组织和细胞中circ_0000502呈高表达,miR-1231呈低表达。沉默circ_0000502或过表达miR-1231降低了TPC-1细胞增殖活力、迁移、侵袭细胞数,抑制PCNA、MMP-2、MMP-9、Bcl-2蛋白表达,促进细胞凋亡和Bax蛋白表达(P<0.05)。circ_0000502靶向调控miR-1231表达(P<0.05),下调miR-1231表达逆转了沉默circ_0000502对TPC-1细胞增殖、迁移、侵袭和凋亡的影响(P<0.05)。 结论沉默circ_0000502通过靶向上调miR-1231调控TPC-1细胞的增殖、迁移、侵袭和凋亡。 |

AimTo investigate the effects of circ_0000502 on the proliferation, migration, invasion and apoptosis of TPC-1 cells in papillary thyroid cancer. Methods26 cases of papillary thyroid cancer tissues and corresponding paracancer tissues were selected. TPC-1 cells were divided into NC group, si-NC group, si-circ_0000502 group, miR-NC group, miR-1231 group, si-circ_0000502+anti-miR-NC group and si-circ_0000502+anti-miR-1231 group. qRT-PCR was used to detect the expression levels of circ_0000502 and miR-1231. MTT was used to detect cell viability. Cell migration and invasion were detected by Transwell system. apoptosis was detected by flow cytometry. The expressions of proliferating cell nuclear antigen (PCNA), matrix metalloproteinase (MMP)-2, MMP-9, B cell lymphoblastoma-2 (Bcl-2) and Bcl-2-associated X protein (Bax) were detected by Western blotting. The targeting relationship between circ_0000502 and miR-1231 was detected by dual-luciferase reporter gene assay. ResultsThe expression levels of circ_0000502 was high and the expression of miR-1231 was low in papillary thyroid cancer tissues and cells. Silencing circle_ 0000502 or overexpression of miR-1231 reduced the proliferative vitality, migration, and invasion number of TPC-1 cells, inhibited the expression of PCNA, MMP-2, MMP-9, and Bcl-2 proteins, promoted cell apoptosis, and Bax protein expression (P<0.05). Downregulation of miR-1231 expression reversed the effects of silencing circ_0000502 on TPC-1 cell proliferation, migration, invasion and apoptosis (P<0.05). ConclusionSilencing circ_0000502 regulates the proliferation, migration, invasion and apoptosis of TPC-1 cells by targeting up-regulation of miR-1231. |
