      目的探究褪黑素在人血管平滑肌细胞(VSMC)巨噬样表型转化中的作用。 方法VSMC分为对照组、胆固醇组(水溶性胆固醇)、褪黑素组(水溶性胆固醇+褪黑素)。水溶性胆固醇诱导VSMC建立巨噬样表型转化模型。定量PCR检测VSMC巨噬样表型相关基因表达水平;油红O染色观察各组VSMC脂质吞噬能力;Edu染色观察各组VSMC增殖情况;细胞划痕实验观察VSMC迁移情况。 结果10 mg/L水溶性胆固醇显著降低平滑肌收缩表型相关标志基因α-actin和α-tropomyosin mRNA表达,并显著升高巨噬细胞相关标志基因CD68和MAC2 mRNA表达,诱导VSMC巨噬样表型转化。0.5 μmol/L褪黑素可抑制水溶性胆固醇引起的VSMC脂质蓄积和细胞增殖,但对VSMC迁移无影响。 结论褪黑素抑制VSMC的巨噬样表型转化。

      AimTo establish a macrophage-like phenotypic transformation model of human vascular smooth muscle cells (VSMC) induced by water-soluble cholesterol, and to explore the role of melatonin in this process. MethodsVSMC is divided into a control group, a cholesterol group (water-soluble cholesterol), and a melatonin group (water-soluble cholesterol+melatonin). Establishment of a macrophage like phenotype transformation model in VSMC induced by water-soluble cholesterol. The expression of macrophage-like phenotypic related genes in VSMC was detected by quantitative PCR. Oil red O staining experiment was conducted to observe the effect of melatonin on the lipid accumulation of VSMC induced by water-soluble cholesterol. Edu staining was used to observe the effect of melatonin on the proliferation of VSMC induced by water-soluble cholesterol. The effect of melatonin on VSMC migration induced by water soluble cholesterol was evaluated by cell scratch assay. ResultsThe mRNA expression of smooth musclecontraction phenotype related markers, α-actin and α-tropomyosin,were significantly decreased after 10 mg/L water-soluble cholesterol treatment, while the mRNA expression macrophage related markers,CD68 and MAC2,were significantly increased after treated by 10 mg/L water-soluble cholesterol, suggesting the transformation of VSMC to a macrophage-like phenotype. In addition, 0.5 μmol/L melatonin inhibited lipid accumulation and proliferation of VSMC induced by water-soluble cholesterol, but had no effect on VSMC migration. ConclusionMelatonin inhibits the macrophage-like phenotype transformation of VSMC.
