      目的采用网络药理学方法探讨膜肾协定方治疗特发性膜性肾病(IMN)的分子机制。 方法应用BATMAN-TCM、TCMSP、TCMID、Swiss Target Prediction、CNKI等数据库收集膜肾协定方相关成分及靶点。应用GeneCards数据库和CTD数据库筛选IMN的靶点;通过在线Venn获得复方-疾病的共同靶点。使用STRING数据库在线预测共同靶点的相互作用,并使用R studio的cluster Profiler包对所筛选靶点进行GO和KEGG通路分析。通过Cytoscape软件绘制膜肾协定方成分调节的网络图。 结果发现膜肾协定方中34个成分,通过作用于PIK3CB、MAPK14、MAPK8、MAPK11等靶点,经Hepatitis B、AGE-RAGE信号通路、cAMP信号通路、FoxO信号通路等177条通路参与调节细胞凋亡、脂质代谢、氧化应激等进程来治疗特发性膜性肾病。 结论膜肾协定方通过多成分作用多靶点,调节多条信号通路发挥对特发性膜性肾病的治疗作用。

      AimTo investigate the molecular mechanism of moshen decoction in the treatment of idiopathic membranous nephropathy (IMN) by network pharmacology. MethodsRelevant components and targets of moshen decoction were collected from BATAC-TCM, TCMSP, TCMID, Swiss Target Prediction, CNKI and other databases. GeneCards database and CTD database were used to screen IMN targets. Compound-common targets of diseases were obtained via online Venn. STRING database was used to predict the interaction of common targets online, and the cluster Profiler package of R studio was used to analyze the GO and KEGG paths for the selected targets. The network diagram of component regulation of moshen decoction was mapped by Cytoscape software. Results34 components of moshen decoction were found. By acting on PIK3CB, MAPK14, MAPK8, MAPK11 and other targets, there were 177 pathways, such as Hepatitis B pathway, AGE-RAGE signaling pathway, cAMP signaling pathway and FoxO signaling pathway, involved in the regulation of apoptosis, lipid metabolism, oxidative stress and other processes to treat idiopathic membranous nephropathy. ConclusionThe moshen decoction exerts its therapeutic effect on idiopathic membranous nephropathy through regulating multiple targets and signaling pathways.
