目的分析泛素羧基末端水解酶37(UCH37)、膜相关RINpCH 8(MARCH8)对宫颈癌细胞增殖、迁移、侵袭的影响。 方法将Hela细胞分为si-NC组、si-UCH37组和si-MARCH8组,利用siRNA技术在Hela细胞中沉默UCH37、MARCH8基因。CCK-8法检测细胞增殖能力。Transwell检测细胞迁移和侵袭能力。qRT-PCR检测细胞UCH37、MARCH8 mRNA表达水平。Western blotting检测细胞UCH37、MARCH8蛋白水平。 结果宫颈癌Hela细胞UCH37、MARCH8 mRNA和蛋白水平明显高于人正常宫颈上皮HUCEC细胞(P<0.05)。与si-NC组比较,si-UCH37组UCH37和si-MARCH8组MARCH8的mRNA和蛋白水平均降低;si-UCH37组和si-MARCH8组细胞增殖活性、迁移细胞数量、侵袭细胞数量均减少(P<0.05)。 结论UCH37和MARCH8在宫颈癌细胞中高表达,干扰其表达后可抑制宫颈癌细胞增殖、迁移和侵袭。 |

AimTo analyze the effects of ubiquitin carboxy-terminal hydrolase 37 (UCH37) and membrane-associated RING-CH 8 (MARCH8) on proliferation, migration and invasion of cervical cancer cells. MethodsHela cells were divided into si-NC group, si-UCH37 group and si-MARCH8 group, in which UCH37 and MARCH8 geneswere silenced by siRNA. The cells proliferation was detected by CCK-8. The cells migration and invasion were detected by Transwell. The expression levels of UCH37 and MARCH8 mRNA were detected by qRT-PCR. The expression levels of UCH37 and MARCH8 proteins were detected by Western blotting. ResultsThe mRNA and proteins of UCH37 and MARCH8 in Hela cells were significantly higher than those in HUCEC cells (P<0.05). Compared with si-NC group, mRNA and protein of UCH37 in si-UCH37 group, and mRNA and protein of MARCH8 in si-MARCH8 group were decreased. Compared with si-NC group, activity of cells proliferation, number of migration and invasion cells in si-UCH37 group and si-MARCH8 group were decreased (P<0.05). ConclusionThe expressions of UCH37and MARCH8 are up-regulated in cervical cancer cells. Interfering their expressions can inhibit the proliferation, migration and invasion of cervical cancer cells. |
