      目的探讨miR-185/YWHAZ轴通过糖酵解对非小细胞肺癌(NSCLC)A549细胞凋亡的影响。 方法糖酵解抑制剂处理后,检测各组糖酵解酶、乳酸、NADH水平和细胞凋亡水平。过表达或敲低miR-185后,过表达或敲低YWHAZ后,均检测细胞乳酸、NADH和细胞凋亡水平。荧光素酶报告实验检测miR-185与YWHAZ mRNA的相互作用。 结果糖酵解抑制剂处理后,糖酵解酶、乳酸、NADH下降,细胞凋亡水平上升(P<0.05)。过表达miR-185时,细胞乳酸、NADH下降,YWHAZ mRNA和蛋白水平下降,细胞凋亡水平上升;敲低miR-185时上述变化逆转(P<0.05)。敲低YWHAZ时,细胞乳酸、NADH下降,细胞凋亡水平上升;过表达YWHAZ时上述变化逆转(P<0.05)。miR-185靶向YWHAZ mRNA的3′端非翻译区。 结论YWHAZ、miR-185分别是促进和抑制糖酵解的关键分子。miR-185靶向YWHAZ mRNA的3′端非翻译区,减少了YWHAZ表达,促进了NSCLC的细胞凋亡。
英文摘要:
      AimTo investigate the effect of miR-185/YWHAZ axis on apoptosis of non-small cell lung cancer (NSCLC) cells through glycolysis. MethodsAfter treatment with glycolytic inhibitors, levels of glycolytic enzymes, lactate,NADH, and cell apoptosis were detected in each group. After overexpression or knockdown of miR-185, and overexpression or knockdown of YWHAZ, the levels of cellular lactate, NADH, and apoptosis were detected. The interaction between miR-185 and YWHAZ mRNA was detected by luciferase report assay. ResultsAfter treatment with glycolysis inhibitor, the glycolytic enzymes, lactate and NADH decreased, and the level of apoptosis increased (P<0.05).When miR-185 was overexpressed, lactic acid and NADH were decreased, YWHAZ mRNA and protein levels were decreased, and apoptosis level was increased, these changes were reversed when miR-185 was knocked down (P<0.05). When YWHAZ was knocked down, lactic acid and NADH decreased and apoptosis level increased, these changes were reversed when YWHAZ was overexpressed (P<0.05). miR-185 targets the 3′-end untranslated region of YWHAZ mRNA. ConclusionYWHAZ and miR-185 are the key molecules in promoting and inhibiting glycolysis, respectively. miR-185 targets the 3′untranslated region of YWHAZ mRNA, reduces YWHAZ expression and promotes apoptosis of NSCLC cells.
