      目的探讨多囊卵巢综合征(PCOS)患者血清miR-222、miR-874-3p和miR-27a水平,并评估其在PCOS中的潜在诊断价值。 方法选取PCOS患者98例为PCOS组,因单纯输卵管不孕或男性因素不孕的妇女98例为对照组。根据体质指数(BMI)、胰岛素抵抗指数(HOMA-IR)和游离雄性激素指数(FAI),将PCOS组分为正常体质量组、超重组和肥胖组;非胰岛素抵抗组和胰岛素抵抗组;高雄激素组和低雄激素组。qRT-PCR检测各组miR-222、miR-874-3p和miR-27a水平,ROC曲线分析miR-222、miR-874-3p和miR-27a在PCOS中的诊断价值。 结果PCOS组BMI、HOMA-IR和FAI及血清miR-222、miR-874-3p、miR-27a均高于对照组(P<0.05)。超重组和肥胖组miR-27a高于正常体质量组,胰岛素抵抗组miR-222和miR-27a高于非胰岛素抵抗组,高雄激素组miR-874-3p高于低雄激素组(P<0.05)。miR-222、miR-874-3p和miR-27a联合诊断PCOS诊断效能最高。 结论PCOS患者血清miR-27a、miR-222和miR-874-3p表达上调,可能与胰岛素抵抗、脂代谢和高雄激素血症有关,三者联合检测有助于PCOS的诊断。

      AimTo investigate the serum levels of miR-222, miR-874-3p and miR-27a in patients with polycystic ovary syndrome (PCOS), and to evaluate their potential diagnostic value in PCOS. Methods98 PCOS patients were selected as PCOS group and 98 women with infertility due to simple tubal or male factors were selected as control group. According to body mass index (BMI), insulin resistance index (HOMA-IR) and free male hormone index (FAI), PCOS group was divided into normal weight group, hyperrecombination group and obesity group. Non-insulin resistant group and insulin resistant group; Hyperandrogen group and hypoandrogen group. miR-222, miR-874-3p and miR-27a in each group were detected by qRT-PCR, and the diagnostic value of miR-222, miR-874-3p and miR-27a in PCOS was analyzed by ROC curve. ResultsBMI, HOMA-IR and FAI and serum miR-222, miR-874-3p and miR-27a in PCOS group were higher than those in control group (P<0.05). The miR-27a of super-recombinant and obese groups was higher than that of normal weight group, the miR-222 and miR-27a of insulin resistance group were higher than those of non-insulin resistance group, and the miR-874-3p of high androgen group was higher than that of low androgen group (P<0.05). miR-222, miR-874-3p and miR-27a have the highest diagnostic efficiency in PCOS. ConclusionThe up-regulated expression of miR-27a, miR-222 and miR-874-3p in serum of PCOS patients may be related to insulin resistance, lipid metabolism and hyperandrogenemia, and the combined detection of the three is helpful for the diagnosis of PCOS.
