      目的探讨环状RNA 0000069(circ_0000069)是否靶向miR-338-3p调控非小细胞肺癌(NSCLC)A549细胞增殖、迁移和侵袭。 方法采用qRT-PCR技术检测NSCLC患者癌组织、癌旁正常组织中circ_0000069和miR-338-3p表达量。采用CCK-8、Transwell实验分析抑制circ_0000069或过表达miR-338-3p对A549细胞增殖活力、迁移和侵袭的影响。使用荧光素酶酶报告实验和qRT-PCR分析circ_0000069和miR-338-3p之间的相互作用。 结果NSCLC组织中circ_0000069表达量较癌旁组织升高(P<0.05),而miR-338-3p表达量显著降低(P<0.05)。抑制circ_0000069或过表达miR-338-3p后A549细胞活力、迁移和侵袭细胞数显著降低(P<0.05)。miR-338-3p是circ_0000069的靶基因,circ_0000069靶向负调控miR-338-3p表达。干扰miR-338-3p表达显著减弱circ_0000069对A549细胞增殖、迁移和侵袭的抑制(P<0.05)。 结论抑制circ_0000069通过上调miR-338-3p抑制NSCLC细胞A549增殖、迁移和侵袭。

      AimTo investigate whether circular RNA 0000069 (circ_0000069) targets miR-338-3p to regulate the proliferation, migration and invasion of non-small cell lung cancer (NSCLC) A549 cells. MethodsqRT-PCR technology was used to measure the expression of circ_0000069 and miR-338-3p in cancer tissues and normal adjacent tissues of NSCLC patients. Cell counting kit method and Transwell experiment were used to analyze the impacts of inhibition of circ_0000069 or overexpression of miR-338-3p on the proliferation, migration and invasion of A549 cells. The luciferase enzyme reporter experiment and qRT-PCR were used to analyze the interaction between circ_0000069 and miR-338-3p. ResultsThe expression of circ_0000069 in NSCLC tissue was obviously increased (P<0.05), while the expression of miR-338-3p was obviously decreased than the adjacent tissues (P<0.05). After inhibiting circ_0000069 or overexpressing miR-338-3p, the viability, migration and invasion of A549 cells were obviously reduced (P<0.05). MiR-338-3p was a target gene of circ_0000069, and circ_0000069 targeted and negatively regulated the expression of miR-338-3p. Inhibition of miR-338-3p expression obviously attenuated the impacts of inhibition of circ_0000069 on the proliferation, migration and invasion of A549 cells (P<0.05). ConclusionInhibition of circ_0000069 can inhibit the proliferation, migration and invasion of NSCLC cells A549 by up-regulating miR-338-3p.
