目的基于生物信息学筛选膝关节骨关节炎(OA)的关键基因、信号通路,并分析软骨组织免疫细胞浸润情况。 方法从GEO数据库中下载来自正常人和OA患者的基因数据集,进行差异表达基因筛选,基因本体论、京都基因和基因组百科全书分析,构建蛋白互作网络,并对软骨相关数据集进行免疫细胞浸润分析。 结果筛选出与OA相关的12个关键基因及86条关键信号通路。免疫浸润结果显示,OA软骨组织与正常软骨比较,其活化CD8T记忆细胞、CD56+NK细胞、髓系来源抑制性细胞、1型辅助型T细胞4种免疫细胞含量差异有统计学意义(P<0.05)。 结论筛选出12个关键基因和4种免疫细胞,其可能参与了OA的进展。 |

AimTo screen key genes, signaling pathways and analyze immune cell infiltration of cartilage tissue in knee osteoarthritis (OA) based on bioinformatics. MethodsGene datasets from normal subjects and OA patients were downloaded from the GEO database for differentially expressed gene screening, gene ontology enrichment, kyoto encyclopedia of genes and genomes analysis, protein-protein interaction network construction, and immune cell infiltration analysis of cartilage-related datasets. Results12 key genes and 86 key signaling pathways related to OA were selected. The results of immune infiltration showed that there was a statistically significant difference in the content of activated CD8T memory cells, CD56+NK cells, myeloid derived inhibitory cells, and type 1 helper T cells in 4 immune cells in OA cartilage compared to normal cartilage (P<0.05). ConclusionThe 12 key genes and 4 immune cell types screened may be involved in the progression of OA. |
