目的探讨葛根素通过沉默调节蛋白3(Sirt3)/腺苷酸活化蛋白激酶(AMPK)信号通路对奥氮平诱导的大鼠糖脂代谢紊乱的影响。方法SPF级SD雄性大鼠以奥氮平灌胃建立糖脂代谢紊乱模型,给予葛根素、Sirt3抑制剂(3-TYP)及二甲双胍干预,比较干预后糖脂代谢指标及Sirt3/AMPK信号通路蛋白表达情况。结果经葛根素干预后大鼠的Lee's指数、空腹血糖(FBG)、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDLC)下降,高密度脂蛋白胆固醇(HDLC)、Sirt3蛋白表达及p-AMPK/AMPK、p-ACC/ACC均升高,联合3-TYP干预后上述指标变化程度降低(P<0.05)。结论葛根素能改善糖脂代谢紊乱大鼠的代谢指标,其机制可能与调节Sirt3蛋白表达和激活AMPK和ACC磷酸化活性有关。 |

To investigate the effect of puerarin on olanzapine-induced glucose and lipid metabolism disorders in rats via the sirt3 (Sirt3)/adenylate-activated protein kinase (AMPK) signaling pathway. MethodsSPF SD male rats were given olanzapine to establish a model of glucose and lipid metabolism disorder, and were given puerarin, Sirt3 inhibitor (3-TYP) and metformin to intervene. Glycolipid metabolism indexes and Sirt3/AMPK signaling pathway protein expression were compared after intervention. ResultsThe Lee's index, fasting blood glucose (FBG), total cholesterol (TC), triglyceride (TG) and low-density lipoprotein cholesterol (LDLC) of rats were decreased after puerarin intervention, High-density lipoprotein cholesterol (HDLC), Sirt3 protein expression, p-AMPK/AMPK, p-ACC/ACC were all increased, and the changes of the above indexes were decreased after combined with 3-TYP intervention(P<0.05).ConclusionPuerarin can improve the metabolic indexes of rats with glucose and lipid metabolism disorder, and its mechanism may be related to the regulation of Sirt3 protein expression and activation of AMPK and ACC phosphorylation activities. |
