      目的探讨氯胍对肝癌细胞Hep-G2及SMMC-7721增殖、集落形成和迁移的影响及可能机制。方法用不同浓度的氯胍分别处理Hep-G2及SMMC-7721细胞。采用MTT法检测细胞增殖情况,克隆形成实验检测细胞集落形成能力,Transwell检测细胞迁移能力,Western blotting检测脂肪酸合成酶(FASN)及调节元件结合蛋白(SREBP-1c)的表达情况。结果氯胍抑制Hep-G2及SMMC-7721的增殖、集落形成及迁移能力;Western blotting显示氯胍下调FASN及SREBP-1c的表达。结论氯胍能抑制肝癌细胞Hep-G2及SMMC-7721增殖、集落形成及迁移,其作用机制可能与下调FASN/SREBP-1c通路有关。

      To investigate the effects of Proguanil on proliferation, colony formation and migration of hepatocellular carcinoma cells Hep-G2 and SMMC-7721. MethodsHep-G2 and SMMC-7721 cells were treated with different concentrations of Proguanil. Cell proliferation was detected by MTT method, colony forming ability was detected by clone formation experiment, cell migration ability was detected by Transwell, and the expression of fatty acid synthase (FASN) and sterol regulatory element-binding protein-1c (SREBP-1c) was detected by Western blotting. ResultsProguanil inhibited the proliferation, colony forming ability and migration of Hep-G2 and SMMC-7721; Western blotting showed that proguanil down-regulated the expression of FASN and SREBP-1c. ConclusionProguanil can inhibit the proliferation, colony formation and migration of liver cancer cells Hep-G2 and SMMC-7721, and its mechanism may be related to down-regulation of the FASN/SREBP-1c pathway.
