      目的探讨迷迭香酸对高糖诱导下人视网膜血管内皮细胞迁移、凋亡的作用。方法将人视网膜血管内皮细胞按培养方案分为对照组、迷迭香酸组、高糖组、联合组。Transwell法观察各组细胞迁移能力,流式细胞术检测各组细胞凋亡水平,ELISA检测各组白细胞介素-6(IL-6)、白细胞介素-8(IL-8)与肿瘤坏死因子-α(TNF-α)的含量。Western blotting法与qRT-PCR技术检测各组细胞中血管内皮生长因子(VEGF)与凋亡相关蛋白Caspase-3、B淋巴细胞瘤-2(Bcl-2)、Bcl-2相关蛋白X(BAX)的表达水平。结果迷迭香酸组与对照组各项指标差异均无显著性(P>0.05)。与对照组比较,高糖组人视网膜血管内皮细胞迁移能力及细胞凋亡水平显著增加,炎症指标IL-6、IL-8与TNF-α的含量增加,VEGF、Caspase-3、BAX蛋白及mRNA水平增加,Bcl-2蛋白及mRNA水平降低(P<0.05)。与高糖组比较,联合组迁移细胞数量、细胞凋亡水平、IL-6、IL-8与TNF-α的含量、VEGF、Caspase-3与BAX的蛋白及mRNA水平降低,Bcl-2蛋白及mRNA水平增加(P<0.05)。结论迷迭香酸可抑制高糖环境下人视网膜血管内皮细胞的迁移及凋亡状态,其机制可能与调控VEGF有关。
英文摘要:
      To investigate the effect of rosmarinic acid on migration and apoptosis of human retinal vascular endothelial cells induced by high glucose. MethodsHuman retinal vascular endothelial cells were divided into control group, rosmarinic acid group, high glucose group and combined group according to the culture scheme. The cell migration ability of each group was observed by Transwell test, the apoptosis level of each group was detected by flow cytometry, and the interleukin-6(IL-6), interleukin-8(IL-8) and tumor necrosis factor-α(TNF-α) were detected by ELISA.The expression levels of vascular endothelial growth factor(VEGF) and apoptosis-related proteins Caspase-3, B-cell lymphoma-2(Bcl-2) and Bcl-2-associated X(BAX) in each group were detected by Western blotting and qRT-PCR. ResultsCompared with the control group, there were no significant differences between rosmarinic acid group and control group (P>0.05). Compared with the control group, the migration ability and apoptosis level of cells in high glucose group were significantly increased, the contents of inflammatory indicators IL-6, IL-8 and TNF-α were increased, the protein and mRNA levels of VEGF, BAX and Caspase-3 were increased, and the protein and mRNA levels of Bcl-2 were decreased (P<0.05). Compared with the high glucose group, the number of migrated cells, apoptosis level, IL-6, IL-8 and TNF-α contents, protein and mRNA levels of VEGF, BAX and Caspase-3 were significantly decreased in the combined group, while protein and mRNA levels of Bcl-2 were significantly increased(P<0.05). ConclusionRosmarinic acid can inhibit the migration and apoptosis of human retinal vascular endothelial cells in high glucose environment, which may be related to the regulation of VEGF signaling pathway.
