      目的探究银杏叶提取物对右美托咪定麻醉小鼠催醒的作用。方法32只昆明种小鼠随机均分为对照组、阿替美唑组、低剂量组及高剂量组。各组小鼠腹腔注射1 mg/kg右美托咪定,90 min后分别注射2 mg/kg阿替美唑、50 mg/kg(低剂量)及100 mg/kg(高剂量)银杏叶提取物,评估小鼠镇静、镇痛水平,并记录苏醒时间。检测各组脑组织中Ca2+、Mg2+-ATP酶、一氧化氮(NO)、一氧化氮合酶(NOS)、鸟苷酸环化酶(cGMP)活性、Orexin A及Orexin B蛋白表达。结果麻醉给药95、120及180 min时,与对照组比较,阿替美唑组和高剂量组镇静、镇痛评分均降低(P<0.05),且麻醉给药180 min时,阿替美唑组和低剂量组镇静、镇痛评分比较,差异有统计学意义(P<0.05);与对照组比较,阿替美唑组和高剂量组苏醒时间缩短(P<0.05),且阿替美唑组苏醒时间短于高剂量组(P<0.05);与对照组比较,阿替美唑组和高剂量组Ca2+、Mg2+-ATP酶活性、NO、NOS、cGMP活性、OrexinA及OrexinB蛋白表达升高(P<0.05)。结论银杏叶提取物对右美托咪定麻醉小鼠具有催醒作用,可能与提高Ca2+、Mg2+-ATP酶活性、促进NO/cGMP信号转导和Orexin系统有关。

      To explore the waking effect of ginkgo biloba extract on mice undergoing dexmedetomidine anesthesia. MethodsA total of 32 Kunming mice were randomly divided into control group, atipamezole group, low-dose and high-dose ginkgo biloba extract groups. Mice in each group were injected intraperitoneally with 1 mg/kg dexmedetomidine, 90 min later, they were injected with 2 mg/kg atipamezole, 50 mg/kg and 100 mg/kg ginkgo biloba extract, respectively, and the levels of sedation and analgesia were assessed, and the awake time was recorded. Mice in each group were decapitated to detect Ca2+, Mg2+-adenosine triphosphate (ATP) enzyme, nitric oxide (NO), nitric oxide synthase (NOS),guanylate cyclase (CGMP) activities, Orexin A and Orexin B in brain tissues. ResultsAt 95,120 and 180 min after anesthesia,scores of sedation and analgesia were decreased in atipamezole group and high-dose ginkgo biloba extract group compared with control group(P<0.05). At 180 min after anesthesia, difference in scores of sedation and analgesia between atipamezole group and low-dose ginkgo biloba extract group was statistically significant (P<0.05).Compared with control group, awake time was significantly shorter in atipamezole group and high-dose ginkgo biloba extract group (P<0.05), which was shorter in atipamezole group than high-dose ginkgo biloba extract group (P<0.05). Compared with control group, Ca2+, Mg2+-ATP enzyme activity, NO, NOS activity, expressions of cGMP, OrexinA and OrexinB were significantly increased in atipamezole group and high-dose ginkgo biloba extract group (P<0.05). ConclusionThere is waking effect of ginkgo biloba extract on mice undergoing dexmedetomidine anesthesia, which may be related to increasing Ca2+and Mg2+-ATP enzyme activity, promoting NO/cGMP signal transduction and Orexin system.
