      目的分析奥拉西坦(Oxi)联合重复经颅磁刺激(rTMS)减轻脑缺血再灌注损伤(I/RI)大鼠缺血侧脑皮质神经元氧化应激损伤的机制。方法60只大鼠随机分为对照组、I/RI组、Oxi组、rTMS组和Oxi+rTMS组。比较各组大鼠神经体征缺失评分(NDS)、脑梗死体积和神经元细胞凋亡率,检测缺血侧脑皮质组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)、B细胞淋巴瘤2(Bcl-2)、Bcl-2相关X蛋白(Bax)、半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)、活化的Caspase-3(Cleaved Caspase-3)、核因子E2相关因子2(Nrf2)、血红素加氧酶(HO-1)和醌氧化还原酶1(NQO-1)蛋白表达。结果与对照组比较,I/RI组NDS评分和神经元细胞凋亡率升高,SOD和GSH-Px水平下降,MDA水平、Nrf2、HO-1、NQO-1、Cleaved Caspase-3/Caspase-3和Bax/Bcl-2蛋白表达升高(P<0.05)。与I/RI组比较,Oxi组、rTMS组和Oxi+rTMS组NDS评分、脑梗死体积和神经元细胞凋亡率下降,SOD和GSH-Px水平、Nrf2、HO-1、NQO-1蛋白表达升高,MDA水平、Cleaved Caspase-3/Caspase-3和Bax/Bcl-2蛋白表达下降(P<0.05);与Oxi组和rTMS组比较,Oxi+rTMS组上述改变更为显著(P<0.05)。结论Oxi联合rTMS可降低脑I/RI大鼠脑组织氧化应激水平,抑制缺血侧脑皮质神经元细胞凋亡,减轻脑组织氧化应激损伤。

      To analyze the mechanism of oxiracetam combined with repeated transcranial magnetic stimulation (rTMS) in alleviating oxidative stress injury of cerebral cortical neurons on the ischemic side of cerebral ischemia/reperfusion injury (I/RI) in rats. MethodsSixty SD rats were randomly divided into sham operation group, I/RI group, Oxi group, rTMS group and Oxi+rTMS group. Neurologic sign loss score (NDS), cerebral infarction volume and neuronal cell apoptosis were compared. The levels of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and malondialdehyde (MDA), B-cell lymphoma 2 (Bcl-2), Bcl-2 related X protein (Bax), cysteine aspartic protease 3 (Caspase-3), Cleaved Caspase-3, nuclear factor E2 related factor 2 (Nrf2), heme oxygenase (HO-1) and quinone oxidoreductase 1 (NQO-1) in cerebral cortex tissue onthe ischemic side were detected. ResultsCompared with sham operation group, NDS and neuronal apoptosis rate were increased in I/RI group, levels of SOD and GSH-Px were decreased, MDA, Nrf2, HO-1, NQO-1, Cleaved Caspase-3/Caspase-3 and Bax/Bcl-2 were increased (P<0.05). Compared with I/RI group, NDS, cerebral infarction volume and neuronal apoptosis rate were decreased in Oxi group, rTMS group and Oxi+rTMS group, levels of SOD, GSH-Px, Nrf2, HO-1 and NQO-1 were increased, MDA, Cleaved Caspase-3/Caspase-3 and Bax/Bcl-2 were decreased (P<0.05). Compared with Oxi group and rTMS group, the above changes were more significant in Oxi+rTMS group (P<0.05). ConclusionOxi combined with rTMS can reduce the level of oxidative stress in brain tissue of rats with cerebral I/RI, inhibit the apoptosis of cerebral cortex neurons on the ischemic side, and alleviate the injury of oxidative stress in brain tissue.
