      目的探析长链非编码RNA(lncRNA)FOXCUT和FOXC1在结直肠癌中的表达及其相关性。方法收集75例结直肠癌组织及癌旁正常组织标本,采用免疫组织化学、蛋白印迹法检测FOXC1表达水平,Real Time PCR检测FOXCUT mRNA和FOXC1 mRNA表达,并分析其相关性及lncRNA FOXCUT与结直肠癌患者临床病理特征的关系。 结果结直肠癌组织中FOXC1表达和lncRNA FOXCUT高表达率均高于癌旁正常组织(P<0.05)。lncRNA FOXCUT表达与结直肠癌TNM分期、AJCC分期、分化程度、远端转移、淋巴结转移等病理特征相关(P<0.05)。结直肠癌组织FOXC1 mRNA、FOXCUT mRNA表达水平高于癌旁正常组织(P<0.05)。经相关性分析,结直肠癌组织FOXC1 mRNA表达与FOXCUT mRNA表达呈正相关(r=0.517,P<0.05)。 结论FOXC1与FOXCUT在结直肠癌组织中呈高表达,且FOXC1 mRNA与FOXCUT mRNA表达呈正相关,推测两者可能相互作用调控肿瘤细胞增殖及侵袭。
英文摘要:
      To analyze the expression and correlation of lncRNA FOXCUT and FOXC1 in colorectal cancer. MethodsCollect 75 cases of colorectal cancer primary lesion tissue specimens and paired normal tissue specimens adjacent to the cancer in our hospital. Immunohistochemistry and Western blotting were used to detect FOXC1 expression levels. Real Time PCR was used to detect FOXCUT mRNA and FOXC1 mRNA in colorectal cancer symptoms.Analyze the correlation and explore the expression characteristics of lncRNA FOXCUT in the clinicopathological characteristics of patients with coloctal cancer. ResultsThe expression of FOXC1 and the high expression rate of lncRNA FOXCUT in colorectal cancer tissue were significantly higher than those in adjacent normal tissues (P<0.05). The expression of lncRNA FOXCUT was correlated with the pathological characteristics of colorectal cancer such as T staging, AJCC staging, degree of differentiation, distant metastasis, and lymph node metastasis (P<0.05). The expression levels of FOXC1 mRNA and FOXCUT mRNA in colorectal cancer tissues were significantly higher than those in adjacent normal tissues (P<0.05). After correlation analysis, the expression of FOXC1 mRNA in colorectal cancer tissue was positively correlated with FOXCUT mRNA (r=0.517, P<0.05). ConclusionFOXC1 and FOXCUT are highly expressed in colorectal cancer cells, and FOXC1 mRNA expression is positively correlated with FOXCUT mRNA, and there is an interaction to regulate tumor cell proliferation and invasion.
