目的研究Hedgehog(Hh)信号通路相关蛋白(Shh、Ptch和Gli1)在前列腺癌患者中的表达及其与疾病预后的关系。 方法取90例手术治疗的前列腺癌患者肿瘤组织及对应的癌旁正常组织标本,采用免疫组化法测定Shh、Ptch和Gli1表达,分析其与临床病理特征的相关性,采用Cox回归模型研究前列腺癌患者术后复发转移的危险因素,并利用Kaplan-Meier生存曲线分析Shh、Ptch和Gli1与无进展生存期的关系。 结果前列腺癌组织中Shh、Ptch和Gli1蛋白表达水平显著高于癌旁正常组织(P<0.05)。不同Shh、Ptch和Gli1蛋白表达的前列腺癌患者的临床分期、Gleason评分、合并骨转移的分布差异具有统计学意义(P<0.05)。Cox回归分析显示,Hh信号通路相关蛋白表达阳性是术后复发转移的独立危险因素(P<0.05)。Kaplan-Meier生存曲线显示,Hh信号通路相关蛋白表达与前列腺癌患者无进展生存期呈负相关(P<0.05)。 结论前列腺癌患者肿瘤组织中Hh信号通路相关蛋白表达与肿瘤分化程度密切相关,对前列腺癌患者预后评估具有一定价值。 |

To investigate the expression and significance of Hedgehog (Hh) signaling pathways related proteins (Shh, Ptch and Gli1), and analyze their relationships with the severity and prognosis in prostate cancer. Methods90 cases of primary prostate cancer patients who underwent surgical treatment were sampled from tumor tissues and corresponding adjacent tissues, expressions of Hh signaling pathway related proteins were determined by immunohistochemistry, the correlation analysis of expressions of Shh, Ptch, Gli1 proteins and clinicopathological characteristics was analyzed. Cox regression analysis was used to analyze the risk factors for postoperative recurrence and metastasis in prostate cancer patients, and Kaplan-Meier survival curve was used to analyze the relationship between the expressions of Hh signaling pathway related proteins and progression-free survival. ResultsThe expression levels of Shh Ptch and Gli1 in prostate cancer tissues were significantly higher than those in adjacent normal tissues (P<0.05). The clinical stage, Gleason score and distribution of combined bone metastasis in prostate cancer patients with different expressions of Shh, Ptch and Gli1 protein was statistically significant (P<0.05). Cox regression analysis results showed that the increased Hh signaling pathway related protein expression was an independent risk factor for postoperative recurrence and metastasis (P<0.05). Kaplan-Meier survival curve showed that expression of Hh signaling pathway related proteins was negatively correlated with progression-free survivalin prostate cancer patients (P<0.05). ConclusionHh signaling pathway related protein expression in tumor tissues of patients with prostate cancer is closely related to the degree of tumor differentiation, which had certain value for prognosis evaluation of patients with prostate cancer. |
