目的探讨葡萄糖调节蛋白78(GRP78)在胃癌中的表达及作用。方法收集不同临床分期和分化程度的胃癌标本48例,癌旁正常胃黏膜组织28例。运用免疫组织化学技术检测胃癌及其转移癌组织中GRP78蛋白的表达;采用MTT及Transwell观察GRP78蛋白对SGC-7901胃癌细胞增殖和迁移能力的影响。结果免疫组织化学结合临床病理分析结果显示:GRP78蛋白位于胞质,且在胃癌组织中GRP78蛋白较正常胃黏膜组织明显增加,其阳性表达率与胃癌的直径、分化、淋巴结转移、分期、复发有关(P<0.05)。MTT及Transwell结果显示:干扰GRP78蛋白表达可抑制SGC-7901胃癌细胞的增殖和迁移能力。结论GRP78蛋白在胃癌组织中高表达,其表达水平与胃癌肿瘤大小、分化程度、淋巴结转移、临床分期及预后有关。 |

To investigate the expression and role of glucose regulated protein 78 (GRP78) in gastric cancer. Methods48 gastric cancer specimens with different clinical stages and degrees of differentiation and 28 adjacent normal gastric mucosa were collected. The expression of GRP78 gene was detected by immunohistochemistry in gastric cancer; The effects of GRP78 gene were observed by MTT and Transwell about the proliferation and migration of SGC-7901 gastric cancer cells. ResultsThe immunohistochemistry combined with clinicopathological analysis showed that GRP78 protein was located in the cytoplasm, and GRP78 protein in gastric cancer was significantly higher than that in normal gastric mucosa. The positive expression rate was related to the diameter, differentiation, lymph node metastasis, stage, recurrence of gastric cancer (P<0.05); MTT and Transwell results showed that interfering with GRP78 gene expression could inhibit the proliferation and migration of SGC-7901 gastric cancer cells. ConclusionsGRP78 gene is highly expressed in gastric cancer tissues. Its expression level is related to tumor size, differentiation, lymph node metastasis, clinical stage and prognosis. |
