      目的观察雾化吸入依达拉奉(EDA)对烟雾吸入性损伤大鼠肺的保护作用。方法将雄性大鼠30只随机均分为对照组、模型组、EDA低剂量组、EDA高剂量组、EDA预防组。对照组不做任何处理,其他各组建立肺损伤模型。计算各组氧合指数(OI)、肺组织含水率及湿干比(W/D);检测血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)含量;测定肺组织丙二醛(MDA)、肺髓过氧化物酶(MPO)、超氧化物歧化酶(SOD)、半胱氨酸蛋白酶-3(Caspase-3)活性及细胞凋亡率;肺组织行苏木精-伊红(HE)染色观察。结果与对照组比较,其余各组肺组织含水率、W/D、TNF-α、IL-6、IL-10、MDA、MPO、Caspase-3及细胞凋亡率水平明显升高(P<0.05),OI及SOD水平降低(P<0.05)。与模型组比较,EDA各组肺组织含水率、W/D、TNF-α、IL-6、MDA、MPO、Caspase-3、细胞凋亡率水平下降(P<0.05),OI、IL-10及SOD水平升高(P<0.05),且EDA预防组变化最显著。镜下EDA各组较模型组肺组织水肿减轻,炎症细胞浸润明显减少。结论依达拉奉能改善烟雾吸入性肺损伤,并呈剂量依赖性,且预防性用药效果更佳。
英文摘要:
      To observe the protective effect of atomization inhalation of edaravone (EDA)on smoke inhalation induced lung injury in rats. MethodsThirty male rats were randomly divided into control group,model group,EDA low-dose group,EDA high-dose group and EDA prevention group. The control group received no treatment, while lung injury model was established in other groups. Oxygenation index(OI), water content and wet to dry ratio(W/D) of lung tissue were calculated. The levels of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), interleukin-10 (IL-10) were tested in serum. The levels of malondialdehyde (MDA), myeloperoxidase (MPO), superoxide dismutase (SOD), cysteine protease 3(Caspase-3), the apoptotic rate were tested in lung tissue homogenate. Part of lung tissue was stained with hematoxylin eosin (HE) and observed under light microscope. ResultsCompared with the control group, the water content, W/D, TNF-α, IL-6, IL-10, MDA, MPO, Caspase-3 and apoptosis rate of lung tissue in other groups were significantly increased (P<0.05), while the OI and SOD levels were decreased (P<0.05). Compared with model group, the water content, W/D, TNF-α, IL-6, MDA, MPO, Caspase-3 and apoptosis rate of lung tissue in EDA groups were significantly decreased (P<0.05), while the levels of OI, IL-10 and SOD were increased (P<0.05), and the above changes obviously in EDA prevention group. Under the microscope, the edema of lung tissue and inflammatory cell infiltration in the EDA groups were significantly reduced compared with the model group. ConclusionEdaravone can improve smoke inhalation lung injury in a dose-dependent manner, and the effect of prophylactic drugs is better.
