      目的探究新疆牧场羊携带微生物多样性及潜在人兽共患病原菌种类,为有效防治人兽共患病提供理论依据。方法选取新疆喀什和石河子市牧场羊群,同期采集羊口咽拭子和肛拭子样品,提取DNA,采用宏基因组二代测序,比对Minikrakenv2数据库,测序结果进行物种注释、多样性和差异性分析,了解两地区羊携带的细菌种类。结果共采集羊口咽拭子72份和肛拭子231份。门水平上,两样品占主导地位的微生物为变形菌门、拟杆菌门和厚壁菌门;属水平上,两样品的优势菌属均不相同,共检出10种主要以食源性传播为主的人兽共患病原菌:大肠弯曲杆菌、空肠弯曲杆菌、大肠杆菌、多杀性巴氏杆菌、鲍曼不动杆菌、沙门氏菌、铜绿假单胞菌、蜡样芽孢杆菌、化脓性链球菌和猪链球菌。经主坐标(PCoA)分析和相似性(ANOSIM)分析,两地区间羊携带细菌和潜在人兽共患病细菌的差异均有统计学意义(P<0.05)。结论新疆地区牧场羊口咽部和肛门部菌群存在多样性及地区差异性,检出的人兽共患病原菌以食源性传播为主,存在食品安全和疾病传播风险。
英文摘要:
      To investigate the bacterial and the species of potential bacterial zoonotic pathogens carried by sheep in large-scale pasture in Xinjiang, China, in order to provide a theoretical basis for effective prevention and control of zoonoses. MethodsBoth oropharyngeal swabs and anal swabs samples were collected at the same time from sheep of the pastures selected in the Kashgar and Shihezi regions of Xinjiang. The genomic DNA in the samples was extracted, macrogenomic sequencing was conducted with second-generation sequencing. The microbial species annotation, similarity and diversity analyses were performed by comparing the minikrakenv2 database to understand the species of bacteria carried by sheep in the two regions. Results72 Oropharyngeal swabs and 231 anal swabs were collected from sheep. At the phylum level, the dominant microorganisms in the two samples were Proteobacteria, Bacteroides and Firmicutes; while at the genus level, the dominant genera were different in both samples. A total of 10 zoonotic pathogens that are mainly food-borne transmission were identified, including Campylobacter coli, Campylobacter jejuni, Escherichia coli, Pasteurella multocida, Acinetobacter baumannii, Salmonella, Pseudomonas guangdongensis, Bacillus cereus, Trueperella pyogenes and Streptococcus porcineus. The results of Principal Coordinates Analysis (PCoA) and Analysis of similarity(ANOSIM)analysis showed that there were statistically significant difference between bacteria carried by sheep in the two regions, and among the potential zoonotic bacterial species in the sheep(P<0.05). ConclusionThe oropharyngeal and anal flora of sheep in Xinjiang pastures are diverse and regionally different, and zoonotic pathogens mainly transmitted by foodborne pathogens were detected, which presented a certain risk of food safety and disease transmission.
