目的分析二烯丙基二硫(DADS)对CD44+胃癌AGS细胞增殖与周期分布的作用,为DADS抑制胃癌作用机制研究提供进一步实验依据。方法经免疫磁珠技术富集人胃癌细胞株AGS中的CD44+细胞,流式细胞术检测CD44+细胞比率。肿瘤球形成、Transwell迁移及侵袭实验验证CD44+细胞的干样特性。MTT、软琼脂集落形成及流式细胞术分析DADS对干样细胞的生长、细胞周期分布的作用。结果免疫磁珠术富集了99.9% CD44+细胞。相较于未分选的AGS组,CD44+AGS组细胞生长2周后成球状态良好,且表现出更强迁移、侵袭能力(P<0.05)。不同质量浓度DADS处理可明显抑制CD44+细胞增殖,且其抑制作用呈质量浓度依赖性(P<0.05);30 mg/L和60 mg/L DADS可抑制CD44+细胞集落形成能力(P<0.05),且可导致细胞周期呈现G2/M期阻滞(P<0.05)。结论DADS可明显削弱CD44+细胞的生长能力,与阻断细胞周期于G2/M期有关。 |

To explore the effect of diallyl disulfide (DADS) on the proliferation and cell cycle distribution of CD44+ gastric cancer stem cell, and provide experimental revealing the anti-cancer mechanism of DADS in gastric cancer.MethodsFirstly, the CD44 positive cell in the human gastric cancer cell line AGS were sorted through immunomagnetic beads technique. The sorting efficiency was analyzed by flow cytometry; the growth characteristics of CD44+ stem-like cell were detected by tumor spheres formation, Transwell migration and invasion experiments; the effects of DADS on the growth and cell cycle distribution of stem like cell was analyzed by MTT, soft agar colony formation and flow cytometry. ResultsThe positive rate of CD44 was 99.9%. Compared with the unsorted cell, the typical tumor spherical colony grew up after 2 weeks culture of CD44 positive cell, and the migration and invasion ability of CD44 positive cell markedly accelerated(P<0.05). DADS at different concentrations significantly inhibited the proliferation of CD44+ cell in a concentration dependent manner(P<0.05). 30 mg/L or 60 mg/LDADS could inhibit the ability of CD44 positive cell colony formation(P<0.05) and lead to G2/M phase arrest of cell cycle(P<0.05). ConclusionDADS can significantly reduce the growth ability of CD44 positive AGS cell, which is related to the arrest of cell cycle in G2/M phase. |
