目的探讨白藜芦醇是否通过激活沉默信息调节因子1(SIRT1)抑制高糖诱导的神经PC12细胞凋亡。 方法PC12细胞用10 μmol/L白藜芦醇预处理30 min,随后加入50 mmol/L葡萄糖溶液处理24 h, qRT-PCR检测SIRT1 mRNA变化,Westren blot检测SIRT1、p-SIRT1、AMPK及p-AMPK表达,Hoechst33258核染色评估细胞凋亡情况,荧光光度法检测细胞活性氧情况,MTT法检测细胞存活率。 结果白藜芦醇预处理,可显著提高细胞存活率(P<0.05),降低高糖(HG)诱导PC12细胞的凋亡率以及活性氧的产生(P<0.05)。此外,Western blot 结果显示HG可显著抑制SIRT1的磷酸化(P<0.05),而这些作用可被白藜芦醇所逆转(P<0.05)。 结论白藜芦醇可能通过激活SIRT1保护 PC12 细胞免受 HG 诱导的凋亡。 |

To investigate whether resveratrol (RESV) antagonizes high glucose (HG) induced apoptosis of PC12 cell by silence information regulator 1 (SIRT1) activation. MethodsPC12 cell were pretreated with 10 μmol/L RESV for 30 min, and then added 50 mmol/L glucose solution for 24 hours, using Quantitative real time-PCR to detect SITR1mRNA, Western blot to measure SIRT1, p-SIRT1, AMPK and p-AMPK protein expression, Hoechst33258 nuclear staining to assess apoptosis, fluorescence photometry to detect reactive oxygen species (ROS) and MTT test to measure cell viability. ResultsResveratrol can increase cell viability and reduce the apoptosis significantly in HG induced PC12 cell(P<0.05), and also decrease ROS(P<0.05). Moreover, HG inhibited SIRT1 phosphorylation could be reversed by resveratrol(P<0.05). ConclusionThese results suggested that RESV could protect PC12 cell from HG induced apoptosis by activating SIRT1. |
