目的建立一种可有效、快速获得的小鼠骨髓间充质干细胞(BMSC)衰老细胞模型,为研究衰老BMSC的生物学特性及功能作准备。 方法造模组采用200 μmol/L过氧化氢(H2O2)处理分离培养的第三代小鼠BMSC 2 h,对照组加入等量PBS处理,两组细胞继续培养72 h。MTT法绘制细胞生长曲线,行SA-β-gal染色和端粒功能失调诱导病灶(TIF)分析,提取细胞RNA,Real-time PCR检测衰老相关基因p16、p21、p53的表达。 结果与对照组比较,H2O2处理后的小鼠BMSC生长缓慢(P<0.05),SA-β-gal染色阳性率较高(P<0.05),TIF阳性细胞较多(P<0.05),且Real-time PCR显示,造模组p16、p21、p53基因表达较高(P<0.05)。 结论通过H2O2处理可以快速且有效地建立BMSC的衰老细胞模型。 |

To establish an effective and rapid aging cell model of mouse bone marrow mesenchymal stem cell (BMSC) and prepare for the study of biological characteristics and function of BMSC. MethodsBMSCs of the third generation mice were treated with 200 μmol/L H2O2 for 2 h in the model group, and the same amount of PBS was added to the control group. The cell of the two groups were cultured for 72 h. MTT method was used to draw the cell growth curve, SA-β-gal staining was used to analyze telomere dysfunction induced foci (TIF), RNA was extracted, and the expressions of p16, p21 and p53 were detected by real-time PCR. ResultsCompared with the control group, the growth of BMSC in H2O2 treated mice was slow (P<0.05), the positive rate of SA-β-gal staining was higher (P<0.05), the number of TIF positive cell was more (P<0.05), and the expression of p16, p21, p53 related genes in the model group was higher (P<0.05). ConclusionAging cell model of BMSC can be established quickly and effectively by H2O2 treatment. |
