      目的探讨miR-365-3p通过周期素依赖性激酶-10(CDK-10)影响口腔鳞癌(OSCC)细胞增殖、凋亡与周期的机制。方法收集在本院就诊的40例口腔鳞癌患者肿瘤组织样本及癌旁正常组织样本,检测其miR-365-3p和CDK-10表达水平。随机将人舌鳞癌细胞系(CAL-27)分为空白组、对照组及miR-365-3p三组。对照组与miR-365-3p组分别转染miR-365-3p NC与miR-365-3p mimic,空白组不进行转染。采用实时荧光定量PCR(qRT-PCR)法检测miR-365-3p表达,CCK-8法及流式细胞术(FCM)检测细胞周期和细胞凋亡。qPCR和Western blot检测CDK-10的表达水平。结果相较于对照组与空白组,miR-365-3p在miR-365-3p组的表达水平显著升高(P<0.05),细胞增殖指数显著降低(P<0.05);细胞凋亡水平显著升高(P<0.05)。转染72 h后,相较于空白组与对照组,miR-365-3p组G1期比例显著降低(P<0.05),S期+G2期比例则显著增加(P<0.05)。转染48 h与72 h后,相较于空白组与对照组CDK-10蛋白在miR-365-3p组的相对表达量显著降低(P<0.05)。结论miR-365-3p在口腔鳞癌细胞中的高表达能够抑制CDK-10表达,从而抑制细胞增殖,调节细胞周期,促进细胞凋亡。

      To explore and analyze the mechanism of miR-365-3p affecting the proliferation, apoptosis and cycle of oral squamous cell carcinoma cells through cyclin-dependent kinase-10 (CDK-10). MethodsCollect 40 patients with oral squamous cell carcinoma who were treated in our hospital, extract the patient’s tumor tissue samples and adjacent normal tissues, detect the expression of miR-365-3p and CDK-10, and randomly select the human tongue cancer cell lines (CAL-27) which were divided into three groups:blank group, control group and miR-365-3p. The control group and the miR-365-3p group were transfected with miR-365-3p NC and miR-365-3p mimic, respectively, and the blank group were not transfected. The expression of miR-365-3p were detected by PCR(qRT-PCR), Cell counting kit-8 (CCK-8) and flow cytometry (FCM) were used to detect cell cycle and apoptosis. The expression of CDK-10 was detected by qPCR and Western blot. ResultsCompared with the control group and the blank group, the expression level of miR-365-3p in the miR-365-3p group was significantly higher (P<0.05), and the cell proliferation index was significantly lower (P<0.05); the apoptosis index was significantly increase (P<0.05). After 72 hours of transfection, compared with the blank group and the control group, the ratio of G1 phase in the miR-365-3p group was significantly reduced (P<0.05), and the ratio of S phase+G2 phase was significantly increased (P<0.05). The relative expression of CDK-10 protein in the experimental group were significantly lower than that in the control group and the blank group at 48 h and 72 h after transfection. ConclusionThe high expression of miR-365-3p in oral scale cancer cells inhibits CDK-10 expression, thereby inhibiting cell proliferation, regulating cell cycles, and promoting apoptosis.
