      目的通过数据挖掘分析TRIM44在结直肠癌中的表达及意义。方法对TCGA和GEO数据库结直肠癌中TRIM44的数据进行R语言脚本分析,并通过GEPIA、TIMER等多个数据库进行了差异表达、生存分析、肿瘤纯度与免疫浸润的关系以及相关基因的共表达和GO与KEGG分析。结果TRIM44在结肠癌中有明显的差异表达(P<0.001),TRIM44高表达者预后较差(P<0.05)。TRIM44在直肠癌中无明显差异表达。GO和KEGG分析结果显示,TRIM44共表达基因富集于调节干细胞多能性信号传导途径等多个通路。进一步免疫浸润分析提示TRIM44与M0和M2免疫细胞基因有明显的相关性。结论TRIM44可通过作用于M0和M2细胞影响CCL2基因表达,进而对结肠癌的发生和发展产生调控作用。TRIM44有望成为结肠癌早期诊断和免疫治疗新靶点。

      To analyze the expression and significance of TRIM44 in colorectal cancer by data-mining.MethodsThis article analyzes the data of TRIM44 in colorectal cancer in TCGA and GEO databases using R script analysis, and conducts differential expression, survival analysis, relationship between tumor cure and immune infiltration, as well as co-expression of related genes through GEPIA, TIMER and other databases and GO and KEGG analysis. ResultsThe results showed that TRIM44 was significantly differentially expressed in colorectal cancer (P<0.001), and those with high expression of TRIM44 had a worse prognosis (P<0.05). There is no significant difference in the expression of TRIM44 in rectal cancer. GO and KEGG analysis results showed that TRIM44 co-expressed genes are enriched in multiple pathways such as the regulation of stem cell pluripotency signaling pathways. Further analysis of immune infiltration indicated that TRIM44 has a significant correlation with M0 and M2 immune cell genes. ConclusionIt is suggested that TRIM44 can affect the CCL2 expression pathway by acting on M0 and M2 cells, thereby regulating the occurrence and development of colorectal cancer. TRIM44 is expected to become a new target for early diagnosis and immunotherapy of colorectal cancer.
