为探讨汉黄芩素对2型糖尿病小鼠视网膜病变的影响,采用酶联免疫法检测小鼠血脂(TC、TG、LDL-C、HDL-C)和空腹胰岛素(FINS)水平;Western blot法和实时荧光定量PCR(qRT-PCR)法分别检测血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)、转化生长因子-β(TGF-β)蛋白及基因水平,分析汉黄芩素对2型糖尿病小鼠视网膜病变的影响。结果显示,汉黄芩素组视网膜VEGF荧光强度较模型组明减弱,TC、TG、LDL-C、FINS含量、VEGF、bFGF、TGF-β的mRNA水平及蛋白表达显著低于模型组,而HDL-C含量明显高于模型组(P<0.05)。汉黄芩素可能通过抑制小鼠糖尿病进程及小鼠视网膜VEGF、bFGF、TGF-β的表达,对小鼠糖尿病视网膜病变起到保护作用。 |

The effect of baicalin on retinopathy of type 2 diabetic mice was explored. In this project, the levels of blood lipids and fasting insulin (FINS) was detected by enzyme-linked immunoassay; the protein and gene levels of VEGF, bFGF and TGF-β were detected by Western blot and Real-Time quantitative PCR (qRT-PCR) respectively, to analyse the effects of baicalin on retinopathy in type 2 diabetic mice. The results showed that the fluorescence intensity of the baicalin group was significantly decreased, the content of TC, TG, FINS,LDL-c and the mRNA level and protein expression of VEGF, bFGF and TGF-beta were significantly lower than that of the model group, while HDL-C content was significantly higher than those in the model group (P<0.05). Baicalin may play a protective role in diabetic retinopathy in mice by inhibiting the progression of diabetes in mice and the expressions of VEGF, bFGF and TGF- beta in mice. |
