探究miRNA-497在麝香保心丸治疗AMI后心室重构中作用。将SD大鼠90只分为假手术组、模型组、miRNA-497激动剂+麝香保心丸组、麝香保心丸组、miRNA-497抑制剂+麝香保心丸组、miRNA-497抑制剂组,每组15只,假手术组仅分离前室间支,不结扎,其余各组采用结扎左冠状动脉前降支法制备AMI大鼠模型。造模成功后进行干预。6周后,miRNA-497激动剂+麝香保心丸组大鼠一般情况、心肌组织病理学改变、左室舒张末压(LVEDP)、左室内压最大上升速率(LV+dp/dtmax)、左室收缩末压(LVESP)、左室内压最大下降速率(LV-dp/dtmax)、急性心肌梗死面积、心肌细胞凋亡率较模型组无明显改善,麝香保心丸组、miRNA-497抑制剂+麝香保心丸组上述指标较模型组有显著改善。麝香保心丸可能通过抑制miRNA-497表达而改善大鼠AMI后心室重构。 |

To investigate the role of microRNA-497 (miRNA-497) in ventricular remodeling of acute myocardial infarction (AMI) treated by Shexiang Baoxin Pill. 90 SD rats were randomly divided into sham operation group, model group, miRNA-497 agonist+Shexiang Baoxin Pill group, Shexiang Baoxin Pill group, miRNA-497 inhibitor+Shexiang Baoxin Pill group, and miRNA-497 inhibitor group, With 15 rats in each group. In the sham operation group, only the anterior interventricular branches were separated without ligation, while rats in other five groups were made into AMI models by ligating the anterior descending branch of the left coronary artery. After the model establishment and 6 weeks of intervention, Comparison was carried for general condition, myocardial histopathological changes, left ventricular end diastolic pressure (LVEDP), maximum rise rate of left ventricular pressure (LV+dP/dtmax), left ventricular end systolic pressure (LVESP), maximum descending rate of left ventricular pressure (LV-dP/dtmax), infarction area and myocardial apoptosis rate, while on significant difference was found between miRNA-497 agonist+Shexiang Baoxin Pill group and model group; Compared with model group, those indexes of Shexiang Baoxin Pill group and miRNA-497 inhibitor+Shexiang Baoxin Pill group were significantly improved, indicating that Shexiang Baoxin Pill may improve ventricular remodeling after AMI in rats by inhibiting miRNA-497 expression. |
