探讨黄芩素是否通过Smad信号通路抑制TGF-β1诱导的肝星状细胞(HSC-T6)活化。用不同浓度黄芩素和/或5 μg/L的转化生长因子β1(TGF-β1)刺激细胞,用免疫细胞化学法测定α-平滑肌肌动蛋白(α-SMA)表达,用Western blot法检测p-Smad 2、p-Smad 3表达。结果显示,TGF-β1可使HSC-T6细胞α-SMA表达增加,黄芩素预处理后,可剂量依赖性地降低α-SMA表达;TGF-β1可使HSC-T6细胞p-Smad 2和p-Smad 3的蛋白水平增加,500 nmol/L黄芩素预处理并未显著降低p-Smad 2和p-Smad 3的蛋白水平。结果说明,黄芩素可抑制TGF-β1诱导的HSC-T6细胞活化,其机制与TGF-β1/Smad信号通路无关。 |

To investigate whether baicalin inhibits TGF-β1-induced activation of HSC-T6 cells via Smad signaling pathway. Hepatic Stellate (HSC-T6) cells were treated with TGF-β1 (5 μg/L) and/or with different concentration of baicalein. Expression of α-SMA was determined using immunocytochemistry, while protein levels of p-Smad 2 and p-Smad 3 by western blot. Results indicated that baicalein decreased α-SMA expression induced by TGF-β1. However, pretreatment with 500 nmol/L baicalein did not significantly change protein levels of p-Smad 2 and p-Smad 3 induced by TGF-β1. In summary, baicalein could suppress TGF-β1-induced activation of HSC-T6 cells independent of TGF-β1/Smad signaling pathway. |
