为探讨慢性前列腺炎患者中核苷酸结合寡聚化结构域 (NOD)样受体家族3 (NLRP3)炎症小体介导的核因子κB(NF-κB)信号通路的表达及变化。采用酶联免疫吸附法(Elisa)检测血清中NLRP3、白细胞介素-1β(IL-1β)、白细胞介素-18(IL-18)、肿瘤坏死因子(TNF-α)、含半胱氨酸的天冬氨酸蛋白水解酶1 (Caspase 1)、Toll样受体4(TLR4)和NF-κB的表达。采用Real-Time PCR检测患者血清中NLRP3、人细胞凋亡相关斑点样蛋白 (PYCARD)、Caspase 1、NF-κB mRNA表达。与健康对照组相比,慢性前列腺炎患者血清炎性因子NLRP3、TLR4和NF-κB含量显著升高,L-Iβ、IL-18、TNF-α和Caspase 1含量显著增高,NLRP3、PYCARD、Caspase 1、NF-κB mRNA含量显著增高。说明检测患者血清中NLRP3/NF-κB信号通路有利于了解患者慢性前列腺炎状态并协助前列腺炎患者的疾病监测。 |

To investigate the expression and changes of the nuclear factor kappa B (NF-κB) signaling pathway mediated by the nucleotide binding oligomerization domain (NOD) receptor family 3 (NLRP3). The serum levels of NLRP3, interleukin-1 beta (IL-1β), interleukin-18 (IL-18), tumor necrosis factor (TNF-α), cysteine, and cysteamine were detected by Elisa. The expression of aspartate protein hydrolase 1 (Caspase 1), Toll like receptor 4 (TLR4) and NF-κB. Real-Time PCR was used to detect the expression of NLRP3, apoptosis related speckle like protein (PYCARD), Caspase 1 and NF-κB mRNA. Compared with the healthy control group, the levels of serum inflammatory factors NLRP3, TLR4 and NF-κB were significantly higher in the patients with chronic prostatitis, and the content of L-Iβ, IL-18, TNF-a and Caspase 1 increased significantly, and the content of NLRP3, PYCARD, Caspase 1 and NF-κB were significantly higher. It is suggested that the detection of serum NLRP3/NF-κB signaling pathway is helpful to understand the state of chronic prostatitis and to assist the disease surveillance of prostatitis patients. |
