      为了研究人脐带间充质干细胞对急性肺损伤大鼠的血清炎性因子及肺组织TRB3 mRNA表达的影响。我们将60只成年SD大鼠分组造模后,治疗组予以气管内滴注人脐带间充质干细胞悬液治疗。用ELISA法测定三组肺组织白介素-6(IL-6)、白介素-10(IL-10)、肿瘤坏死因子-α(TGF-α)水平,实时荧光定量PCR检测肺组织TRB3 mRNA表达量,称量计算大鼠肺组织湿/干质量比。研究发现应用干细胞治疗后,肺组织内IL-6、IL-10、TGF-α水平显著降低,TRB3 mRNA表达量也降低,肺组织湿/干质量降低,水肿明显缓解。说明人脐带间充质干细胞可有效降低急性肺损伤大鼠的血清炎性因子及肺组织TRB3 mRNA表达水平,并减少肺组织湿/干质量比。
英文摘要:
      To investigate the effects of human umbilical cord mesenchymal stem cells on serum inflammatory cytokines and TRB3 mRNA expression in lungs of rats with acute lung injury. After 60 adult SD rats were divided into groups, the treatment group was treated with intratracheal instillation of human umbilical cord mesenchymal stem cell suspension. The levels of interleukins-6 (IL-6), interleukins-10 (IL-10) and tumor necrosis factor-α (TGF-α) in the three groups of lung tissue were measured by ELISA. The expression of TRB3 mRNA in lung tissue was detected by real-time fluorescence quantitative PCR. The wet / dry mass ratio of lung tissue was weighed. The results showed that the levels of IL-6, IL-10 and TGFα in lung tissue were significantly decreased, the expression of TRB3 mRNA was decreased, the wet / dry weight of lung tissue was decreased, and edema was relieved. Human umbilical cord mesenchymal stem cells can effectively reduce serum inflammatory cytokines and lung tissue TRB3 mRNA expression in acute lung injury rats, and reduce the lung wet / dry mass ratio.
