      通过比较妊娠期糖尿病(GDM)孕妇和正常妊娠孕妇胎盘组织中胰岛素样生长因子-1(IGF-1)和胰岛素样生长因子-1受体(IGF-1R)表达,以及新生儿胰岛β细胞功能指数(HOMA-β)和胰岛素抵抗指数(HOMA-IR),从而探讨胎盘组织IGF-1和IGF-1R表达与新生儿HOMA-β和HOMA-IR的相关性。结果显示:GDM组孕妇胎盘组织IGF-1和IGF-1R的mRNA和蛋白表达均显著高于对照组(P<0.05);GDM组孕妇分娩新生儿HOMA-IR显著高于对照组(P<0.05),而HOMA-β明显低于对照组(P<0.05);Spearman分析显示孕妇胎盘组织IGF-1和IGF-1R表达与新生儿HOMA-β呈负相关,而与HOMA-IR呈正相关。说明妊娠期糖尿病孕妇胎盘组织IGF-1和IGF-1R表达增加,导致新生儿胰岛β细胞功能受损和胰岛素抵抗。

      In this paper, the expression of IGF-1 and IGF-1R in placenta tissue of gestational diabetes mellitus and normal pregnant women were compared, the homeostatic model assessment for β-cell function (HOMA-β) and homeostatic model assessment for insulin resistance (HOMA-IR) in newborns were compared as well, in order to investigate the correlation between IGF-1, IGF-1R and HOMA-β, HOMA-IR. Real-time fluorescence quantitative PCR and Western Blot detection showed that:the expression of IGF-1 and IGF-1R in placenta tissue of the GDM group was significantly higher than that in the control group (P<0.05), the HOMA-IR of GDM group was significantly higher than that in the control group (P<0.05), while the HOMA-β was significantly lower than that in the control group (P<0.05). Spearman analysis showed that the expression of IGF-1 and IGF-1R in placenta tissues was negatively correlated with HOMA-β, but positively correlated with HOMA-IR. The results of this study showed that:the expression of IGF-1 and IGF-1R in placenta tissue of gestational diabetes mellitus increased significantly, and leads to impaired β-cell function and insulin resistance in newborns.
