      STGC3基因是一个鼻咽癌相关候选抑癌基因,其对鼻咽癌放疗的影响尚不清楚。本文通过构建STGC3基因高表达CNE2细胞系,探讨STGC3基因高表达对人鼻咽癌CNE2细胞放疗敏感性的影响及其可能的分子机制。成功构建和鉴定STGC3基因高表达CNE2细胞系(CNE2-RTP-hSTGC3-his)后,总剂量4 Gy的6 MV X线照射各组细胞。CCK-8结果显示实验组细胞(CNE2-RTP-hSTGC3-his)的活力较阴性对照组细胞(CNE2-RTP-his)和空白对照组细胞(CNE2)下降(P<0.05);凝胶电泳及质谱分析结果显示SFN蛋白、微管蛋白(TUBA1B)、钙联蛋白(calnexin)在X线照射后的表达增加。同时,与各对照组相比较,实验组细胞中的自噬相关蛋白微管相关蛋白1轻链3(LC3)和SFN蛋白的表达下降(P<0.05),而p62的表达量则升高(P<0.05)。实验表明,上调STGC3基因可能通过抑制SFN的表达而抑制放疗引起的CNE2细胞自噬,从而提高CNE2细胞对放疗的敏感性。

      STGC3 gene is a candidate tumor suppressor gene of Nasopharyngeal carcinoma (NPC), however, the role of STGC3 in NPC radiotherapy is uncertain.This study established the CNE2 cell line with overexpression of STGC3 gene and investigated the influences of STGC3 gene overexpression on radiosensitivity of CNE2 cells and discussed its potential mechanism. After successful expression of STGC3 gene in CNE2 verified by Western blot, the dose of 4Gy of X-ray was used to pretreat the cells. The CCK-8 assay detected that the survival rate of CNE2-RTP-hSTGC3-his cell was decreased after irradiation(P<0.05). The analysis of SDS-PAGE electrophoresis and Protein Mass Spectrometry revealed that the expression of SFN, TUBA1B and calnexin protein were increased after irradiation. The results of Western blot found the expression of SFN protein and LC3 protein were decreased in CNE2-RTP-hSTGC3-his cells(P<0.05) and the expression of p62 protein was increased(P<0.05). Our data suggested that the overexpression of STGC3 gene could enhance the radiosensitivity via the inhibition of autophagy induced by irradiation in CNE2 cells.
