      目的 观察蛞蝓有效提取物(EL-3)对宫颈癌HeLa细胞裸鼠移植瘤生长的影响。方法制备宫颈癌HeLa细胞裸鼠移植瘤模型,检查EL-3对瘤体积、瘤重及VEGF蛋白表达的影响,评估EL-3在体内治疗宫颈癌的有效性。结果EL-3(10、30和100 mg/kg)实验组的瘤体积显著低于生理盐水组(P<0.05),同时,其移植瘤的瘤重抑制率分别为42.0%、67.0%和83.0%,提示EL-3在体内抗宫颈癌作用较强,且呈剂量依赖性;采用免疫组化检测证实EL-3下调宫颈癌裸鼠移植瘤细胞VEGF蛋白表达。结论EL-3具有抑制宫颈癌HeLa细胞裸鼠移植瘤生长的作用,其机理可能与抑制肿瘤血管生成有关。
英文摘要:
      Objective To determine the effect of EL-3,an effective extraction of limax,on the growth of cervical cancer HeLa cell xenografts in nude mice.MethodsThe model of cervical cancer HeLa cells in nude mice was established,and the effect of EL-3 on the tumor volume,tumor weight and the expression of VEGF protein were examined.The effectiveness of EL-3 in the treatment of cervical cancer in vivo was evaluated.ResultsEL-3 can effectively inhibit the growth of human cervical cancer transplanted tumor in nude mice;tumor volume of 10,0,100 mg/kg dose group of EL-3 were significantly lower than that of normal saline group (P<0.05);simultaneously,the transplanted tumor weight inhibitory rates were 42%,67% and 83% respectively,suggesting that the effect of EL-3 in vivo against cervical cancer is stronger and in a dose-dependent manner.Immunohistochemistry showed that EL-3 could down regulate the expression of VEGF protein of the transplanted tumor tissues of cervical cancer HeLa cells in nude mice.ConclusionEL-3 can inhibit the tumor growth of cervical cancer HeLa cells in nude mice,and the mechanism may be related to the inhibition of tumor angiogenesis.
