      目的 研究重组人促红细胞生成素(rhEPO)是否可以通过调节PI3K/AKT信号通路来改善大鼠脑出血后的神经元损伤。方法SD大鼠构建脑出血(ICH)模型,并给予rhEPO药物治疗,采用TUNEL法和试剂盒检测神经细胞凋亡及凋亡蛋白Caspase 9表达;采用Real-Time PCR法和Western blot法检测PI3K、AKT的基因和蛋白磷酸化水平的表达。结果与ICH非治疗组相比,rhEPO治疗组中神经元凋亡细胞数和Caspase 9活性表达明显降低(P<0.05); rhEPO治疗组PI3K和AKT的蛋白磷酸化水平表达和mRNA表达显著升高 (P<0.05)。结论重组人促红细胞生成素rhEPO具有神经保护作用,可以通过调节PI3K/AKT信号来改善大鼠脑出血后的神经元损伤。
英文摘要:
      Objective To investigate the effect of recombinant human erythropoietin (rhEPO) on neuronal injury after intracerebral hemorrhage in rats and PI3K/ AKT signaling pathway.MethodsSD rats were used to construct intracerebral hemorrhage (ICH) model,and rhEPO was given to therapy rats.The apoptosis of neurons was detected by TUNEL method.The expression of Caspase 9 was detected by assay kit.Real-time PCR and Western blot were used to detect the expression of PI3K and AKT mRNA expression.ResultsCompared with ICH group,the number of apoptotic cells and the expression of Caspase 9 in rhEPO group were significantly decreased,the expression of PI3K and AKT protein phosphorylation and mRNA expression in rhEPO group were significantly higher than those in control group (P <0.05).ConclusionRecombinant human erythropoietin rhEPO obviously has a neuroprotective effect in rats,which could be used to reduce neuronal injury after intracerebral hemorrhage by regulating PI3K/ AKT signaling.
