目的 观察游泳训练对新生大鼠学习记忆和海马神经发生的影响。方法雄性7日龄新生SD大鼠随机分为对照和游泳训练组。训练组大鼠出生后第11天进行无负重游泳训练3周。Morris水迷宫法检测学习记忆能力,BrdU+/NeuN+免疫荧光双标染色分析海马神经发生,Western blot检测海马脑源性神经营养因子(BDNF)蛋白表达。结果对照和游泳训练组大鼠随着训练天数增加,寻找平台潜伏期均明显缩短(均P<0.05);定位航行实验的第二、三和四天,游泳训练组大鼠寻找平台潜伏期较对照组均显著减小(均P<0.05)。游泳训练组大鼠穿越目标次数何时间明显增加(均P<0.05)。与对照相比,游泳训练组大鼠海马齿状回NeuN+/BrdU+阳性数量显著增加(P<0.05),BDNF表达显著增加(P<0.05)。结论游泳训练提高了新生大鼠的学习记忆能力,其机制可能与游泳训练显著上调大鼠海马BDNF的表达促进海马神经发生有关。 |

Objective To observe effect of swimming training on learning and memory and hippocampus neurogenesis in neonatal rats.Methods7-days-old newborn Sprague-Dawley rats were randomly divided into control and swimming training group.From eleventh days after birth,the rats in swimming group were carried on 3-weeks swimming training with no load once a day,60 min every time.Learning and memory was evaluated by Morris water maze.The number of BrdU+/NeuN+ was counted by immunofluorescence double labeling staining.The expression of brain derived neurotrophic factor (BDNF) was tested by Western blot.ResultsThe latency for finding the hidden platform in control and swimming training group was obviously decreased with the training days increasing (all P<0.05).On the second,third and forth day,the latency for finding the hidden platform in swimming training group was significantly decreased compared with control (all P<0.05).Number across platform and the time across platform was significantly increased in swimming training group compared with control (all P<0.05).Number of NeuN+/BrdU+ cells was significantly increased and expression of BDNF was significantly increased in swimming training group compared with control (all P<0.05).ConclusionSwimming training increases capacity of learning and memory in neonatal rats,which mechanism may be related with up-regulation of BDNF expression and increase of neurogenesis in hippocampus of rats. |
