目的 探讨miR-195对BEL-7402/5-FU细胞5-氟尿嘧啶药物敏感性的影响。方法采用qRT-PCR检测BEL-7402细胞与BEL-7402/5-FU细胞中miR-195的表达水平。将miR-195质粒载体瞬时转染至BEL-7402/5-FU细胞;MTT检测细胞药物敏感性的改变。结果相比于BEL-7402细胞,miR-195在BEL-7402/5-FU细胞中表达水平下调;miR-195转染组的细胞增殖抑制率较miRNA阴性对照组及未转染组明显增高。结论MiR-195能够增加BEL-7402/5-FU细胞对5-氟尿嘧啶的药物敏感性,抑制细胞增殖。 |

Objective This study investigated the effect of miR-195 on 5-FU resistance in BEL-7402/5-FU Cells.MethodsqRT-PCR analyzed expression of miR-195 between BEL-7402 and BEL-7402/5-FU cells.MiR-195 was transiently transfected into BEL-7402/5-FU cells.Drug sensitivity of the cells to 5-FU was analyzed by MTT.ResultsThe expression of miR-195 was downregulated in BEL-7402/5-FU cells compared to parental BEL-7402 cells.MTT results showed that miR-195 transfected cells had a higher cell inhibition rate than untreated cells or negative control.ConclusionsMiR-195 could sensitize BEL-7402/5-FU cells to 5-FU and inhibited cell proliferation. |
