      目的 探讨血管紧张素Ⅱ对人脐静脉内皮细胞(HUVECs)中降钙素基因相关肽(CGRP)受体组分的影响。方法以HUVECs作为实验对象,用无血清培养基同步化24 h后,以Ang Ⅱ非处理细胞为对照组,AngⅡ处理组为不同剂量的AngⅡ处理HUVECs 12 h,流式细胞仪检测细胞增殖率和凋亡率。Western Blot检测CGRP三种受体组分,即降钙素受体样受体(CRLR)、降钙素受体组分蛋白(RCP)和受体修饰蛋白(RAMP)1的蛋白表达。结果与对照组相比,在处理组细胞,AngⅡ呈剂量依赖性地抑制HUVECs增殖和诱导细胞凋亡,同时上调CRLR表达(P<0.01)。而对另外两个受体蛋白而言,AngⅡ(0.01~100 nmol/L)呈剂量依赖性的下调HUVECs的RAMP1的表达和RCP表达(P<0.05)。结论AngⅡ影响人脐静脉内皮细胞CGRP受体组分的表达存在差异,可引起人脐静脉内皮细胞CGRP受体功能改变。

      Objective To investigate the effect of angiotensin Ⅱ on the expression of calcitonin gene related peptide (CGRP) in human umbilical vein endothelial cells (HUVECs).MethodsHUVECs was employed as the experimental object,cells were incubated by free serum for 24 h for synchronous cell cycle,then,Ang Ⅱ group cells were incubated with different concentration of angiotensin Ⅱ (Ang Ⅱ),the control cells were incubated by free Ang Ⅱ.The cell proliferation and apoptosis were observed by flow cytometry.Western blot was used to determine the expressions of calcitonin receptor like receptor (CRLR),receptor activity modifying protein-1 (RAMP1) and receptor component protein (RCP).ResultsCompared with the control group,in the treated cells,Ang Ⅱ inhibited the HUVECs proliferation and induced cell apoptosis,and also up-regulated the CRLR expression of HUVECs (P<0.01) as well.In addition to the other two receptor proteins,Ang Ⅱ (0.01~100 nmol/L)down regulated expression of RAMP1 and RCP expression (P<0.05)in a dose-dependent manner.ConclusionAng Ⅱ can affect the expression of CGRP receptor in HUVECs,which can cause the functional changes of CGRP receptor in the cells.
