      目的 观察山银花黄酮类提取物对人肝L-02细胞甘油三酯蓄积的影响,并初步探讨SREBP-1c蛋白及基因表达在其中的作用。 方法 油红O染色初步观察山银花黄酮类提取物对脂变人肝L-02细胞中脂滴形成的影响。然后以脂变人肝L-02细胞为模型,不同浓度(0、5、10、20 mg/L)山银花黄酮类提取物作用于细胞24 h以及10 mg/L山银花黄酮类提取物作用于细胞不同时间(0、12、24、48、72 h),生化试剂盒测定细胞内甘油三酯(TG)的含量,Western印迹法检测细胞内SREBP-1c蛋白表达变化,实时荧光定量PCR检测细胞内SREBP-1c mRNA表达变化。 结果 山银花黄酮类提取物显著降低人肝L-02细胞中脂滴含量,细胞内TG水平下降,SREBP-1c蛋白及基因表达下调,且呈现一定的剂量和时间依赖性。 结论 山银花中黄酮类提取物能引起人肝L-02细胞TG含量下降,且这种作用与SREBP-1c蛋白及基因表达下调有一定关系。

      Objective To observe the effect of lavonoid extract from Flos Lonicerae Confusae on the triglyceride accumulation in human L-02 hepatocyte,and to investigate the effect of the protein and mRNA expression of SREBP-1c in this process. Method The effect of the lavonoid extracted from Flos Lonicerae Confusae on lipid-loaded human L-02 hepatocyte,and the lipid droplet density in cells is observed by oil red O staining.Then the lavonoid extracted from Flos Lonicerae Confusae of different density (0,5,10,20 mg/L for 24 h) and different time (0,12,24,48,72 hours of 10 mg/L)had effects on lipid-loaded human L-02 hepatocyte,the level of triglyceride was dected by using 3-glycerol phosphate oxidase method,and the protein and mRNA expression of SREBP-1c was dected by Western blot and real-time quantitative PCR. Results Flavonoid extract from Lonicerae Confusae had effects onipid-loaded human L-02 hepatocyte,the lipid droplet density decreases in cells,the level of triglyceride decreased,the protein and mRNA expression of SREBP-1c were down-regulated,in a dose and time-dependent manner. Conclusion The lavonoid extract from Lonicerae Confusae can degradete the level of triglyceride in lipid-loaded human L-02 hepatocyte,and the possible mechanism is related to the down-regulation of SREBP-1c expression.
