      目的 探讨微水RNA-122(miR-122)对5-氟尿嘧啶(5-FU)诱导BEL-7402/5-FU细胞凋亡的影响。 方法 构建miR-122和阴性对照质粒瞬时转染至BEL-7402/5-FU细胞;流式细胞术检测细胞凋亡情况;Western blot检测caspase-8,caspase-9和caspase-3蛋白表达水平。 结果 仅用miR-122处理BEL-7402/5-FU细胞时,与未转染组和转染对照组比较,miRNA-122转染组细胞凋亡率增加,用10 μmol/mL 5-FU作用BEL-7402/5-FU细胞24 h后,与未转染组和转染对照组比较,miR-122转染组细胞凋亡率明显增加;与未转染组和转染对照组比较,转染了miRNA-122的BEL-7402/5-FU细胞中caspase-8、caspase-9和caspase-3前体蛋白表达降低,活化的caspase-8、caspase-9和caspase-3蛋白表达升高。 结论 miR-122能够下调BEL-7402/5-FU细胞中caspase-8、caspase-9和caspase-3前体蛋白表达,上调活化的caspase-8、caspase-9和caspase-3蛋白表达,miR-122能促进5-FU诱导的BEL-7402/5-FU细胞凋亡。

      Objective This study deals with the effects of miRNA-122 on BEL-7402/5-FU cell apoptosis induced by 5-fluracil. Method MiR-122 and control miRNA expression vectors were transient transfected into BEL-7402/5-FU cells.Cell apoptotic percentage was detected by flow cytometry.Western blot was used to detect the caspase-8,caspase-9 and caspase-3 protein expression. Result Flow cytometry results demonstrated that miR-122 transfected cells had a higher apoptosis rate than BEL-7402/5-FU cells or control group.Moreover,miR-122 transfected cells had significant move value than BEL-7402/5-FU cells or control group by 10 μmol/ml 5-FU for 24 hours.Western bolt results showed that the expression level of pro-caspase-8,pro-caspase-9 and pro-caspase-3 in miR-122 transfected cells were obviously decreased,while the expression level of active caspase-8,caspase-9 and caspase-3 were obviously increased compared with BEL-7402/5-FU cells or control group. Conclusion miR-122 down-regulate pro-caspase-8,pro-caspase-9 and pro-caspase-3 expressions and up-regulate active caspase-8,caspase-9 and caspase-3 expressions in BEL-7402/5-FU cells.MiR-122 increased the cell apoptosis induced by 5-fluracil.
